| [Background]Kidney stone is a common disease with high prevalence and recurrence rate.The prevalence of kidney stones in Chinese males is 6.5% and in Chinese females is 5.1%.The recurrence rates of calcium oxalate stones are 50% in 10 years.The pathogenesis of nephrolithiasis is not clear.Recently,the involvement of the gut microbiota in the pathogenesis of kidney stone has been hypothesized since the discovery of gut–kidney axis.Due to the serious complications and high recurrence rates,it is necessary to dissect the mechanism that contributes to nephrolithiasis formation with an aim to prevent the recurrence of kidney stone.16SrRNA is a conserved piece of sequence in the bacteria genome.We can acquire complete information of specimen through high-flux 16SrRNA’s metagenomic sequencing and explore the relationship between gut microbiome and nephrolithiasis.[Objective]Comparing the difference of intestinal microecological core bacteria between nephrolithiasis patient and healthy subjects by high-flux 16SrRNA’s metagenomic sequencing technology.[Methods]Collecting the fecal samples of 20 calcium oxalate kidney stones patient(SLs)and healthy subjects(SNs);appraising the genera type of subjects’ DNA in stools bacteria flora through 16SrRNA’s metagenomic sequencing technology;optimizing the quality of the results;building the testing systems of intestinal microflora.Analyzing the different traits of bacteria flora in faeces between calcium oxalate kidney stones patient and healthy subjects through the microbiome composition and the species difference analysis.[Results](1)We obtain the complete DNA information of stools through the high-flux 16SrRNA’s metagenomic sequencing technology;and there is a complete microbial flora structure in intestinal tract of kidney stones patient.(2)There is no difference in intestinal microbiota composition between calcium oxalate kidney stones patient and healthy people.(3)The Oxalobacter abundance was not different between two groups(both low).(4)Compared with SNs,faecal samples from SLs exhibited a significantly lower bacterial representation of genes of lactobacillus and bifidobacterium.[Conclusion](1)16SrRNA’s metagenomic sequencing technology can be taken as a reliable appraisal system to identify intestinal microflora genera.(2)Calcium oxalate kidney stones patients has distinct difference in intestinal microbe core flora.It can be a new treatment of nephrolithiasis to intervention intestinal microbiota. |
