| Objective:To study the expression and clinicopathological characteristics of Circ-SIRT1 in patients with colorectal cancer,compare its difference in normal adjacent tissues.The expression of Circ-SIRT1 was detected in human colon normal epithelial cells and colorectal cancer cells,and the difference in its expression level was explored.Methods:1.Trizol method was used to extract total RNA from colorectal cancer tissues and normal adjacent tissues and their plasma from 30 patients,and the total RNA extracted from the plasma of 30 healthy people in the same way.2.To verify the integrity of the extracted RNA by agarosegel electrophoresis.3.Real-time fluorescent quantitative polymerase chain reaction(qRT-PCR)method was used to detect the expression of Circ-SIRT1.4.Analyze the expression differences of Circ-SIRT1 in various specimens,compare its relationship with clinicopathological staging.5.The expression of Circ-SIRT1 was detected in human colon normal epithelial cells and colorectal cancer cells.Results:The expression of Circ-SIRT1 in colorectal cancer tissues is higher than that in normal adjacent tissues,its expression level in the plasma of colorectal cancer patients is higher than that of healthy people,and is obviously related to tumor differentiation.The expression level of Circ-SIRT1 in colon normal epithelial cells FHC was significantly higher than its expression in colorectal cancer cell lines HCT116 and SW480.Conclusions:1.Circ-SIRT1 is up-regulated in colorectal cancer tissues.2.Circ-SIRT1 is up-regulated in the plasma of patients with colorectal cancer.3.The expression level of Circ-SIRT1 is positively correlated with the degree of tumor differentiation. |
