| Objective To investigate the role of Squalene Synthetase(SQS)in malignant phenotypes of PC-3 and LNCa P cells.To observe the relationship between the antitumor effect of Celastrol in prostate cancer cells and its effect on SQS.Methods Choose 2 kinds of prostate cancer cells: PC-3 and LNCa P.The 2 prostate cancer cells were treated with Celastrol at different times and concentrations.The effect of Celastrol on the proliferation of the two cancer cells were determined by CCK-8 proliferation test.The effects of Celastrol on the migration of the two cancer cells were determined by Transwell migration test and scratch test.Western Blot was used to detect the effect of Celastrol on the expression of squalene synthetase in 2 prostate cancer cells.The SQS gene(also known as FDFT1 gene in human)was disturbed or overexpressed,and the effects of modified and combined with Celastrol treatment on cell proliferation and migration were observed by CCK-8 proliferation test,scratch test and Transwell migration test,respectively.Results Celastrol could inhibit the proliferation and migration of PC-3 and LNCa P prostate cancer cells(P<0.05)in a dose-dependent manner,and reduce the expression of SQS in 2 prostate cancer cells(P<0.05).Interfering with SQS expression could inhibit the proliferation and migration of PC-3 and LNCa P prostate cancer cells(P<0.05).After interfering with SQS,the addition of Celastrol still could inhibit the proliferation and migration of 2 prostate cancer cells(P<0.05).Overexpression of SQS could inhibit the proliferation and migration of PC-3 cells(P<0.05),but there was no significant change in the proliferation and migration ability of LNCa P cells.After SQS overexpression,the addition of Celastrol still could inhibit the proliferation and migration ability of PC-3 cells(P<0.05)Conclusion Celastrol can inhibit the proliferation and migration of PC-3 and LNCa P cells.Celastrol can reduce the expression of SQS in PC-3 and LNCa P cells.SQS does play a role in the malignant behavior of prostate cancer cells.The inhibitory effect of Celastrol on the malignant phenotype of prostate cancer cells is partly dependent on SQS.SQS may have a dual effect in the malignant phenotype of prostate cancer cells. |
