Objective To investigate the expression and clinical significance of Phospholipid hydrogen glutathione peroxidase(PHGPX or Gpx4)and Proliferating Cell Nuclear Antigen(PCNA)in diffuse large B-cell lymphoma(DLBCL).Methods 1.In this study,67 patients with DLBCL were collected from the Department of Pathology of Gansu Provincial people’s Hospital from January 2016 to December 2019.In the same period,20 cases of lymph node reactive hyperplasia were collected as control group.2.Immunohistochemical method was used to detect the positive expression of Gpx4 protein and PCNA protein in 67 cases of DLBCL(lymphoid tumor group)and 20 cases of lymph node reactive hyperplasia(control group),and the relationship between Gpx4 and PCNA and clinicopathological features of the patients was analyzed.3.To analyze the correlation between the expression of Gpx4 and PCNA in DLBCL tissues.Results The positive expression of Gpx4 in DLBCL tissue was yellow and brown granule,mainly in the cell membrane of lymphoma cells and scattered in the nucleus.The positive expression rate of DLBCL was 31.34% in DLBCL tissue;The positive expression of PCNA in DLBCL tissues was brown,mainly in the nucleus,and the positive expression rate in DLBCL tissues was 64.18%.The difference was statistically significant.(P<0.05).In addition,Gpx4 was closely correlated with clinical stage,B symptom and C-reactive protein(C-reactive protein,CRP)in patients with DLBCL(P<0.05);The PCNA was closely related to lactate dehydrogenase level,clinical stage,bone marrow invasion,B symptom and erythrocyte sedimentation rate in patients with DLBCL(P<0.05);while Gpx4 and PCNA were not related to sex,age,β 2-microglobulin and pathological classification of DLBCL patients.Further study showed that there was a positive correlation between the expression of Gpx4 and PCNA in DLBCL tissues(r=0.164;P=0.013).Conclusion1.The expression levels of Gpx4 and PCNA in DLBCL tissues were higher than control group tissues,and were related to tumor stage and B symptoms.2.There has a correlation between the expression of Gpx4 and PCNA in DLBCL tissues.3.The high expression of Gpx4 and PCNA may be involved in the occurrence and development of DLBCL,which may provide a theoretical basis for the pathogenesis and treatment of DLBCL.
