Objective The development of chemoresistance in tumour cells is often accompanied by epithelial-to-mesenchymal transition(EMT),and paired-related homology box 1(PRRX1)can be an inducer that promotes the development o f EMT in tumour cells.Studies have shown that PRRX1 expression levels are significantly increased i n gastric cancer cells compared to gastric paraneoplastic tissues,and increase the invasiveness and metastatic ability of gastric cancer.This study investigated the effect of PRRX1 expression level and resistance of gastric cancer cells to oxaliplatin(L-OHP)to provide research ideas for the prevention and treatment of drug resistance in gastric cancer.Methods 1.In vitro concentration incremental method was used to construct SGC-7901/L-OHP,an oxaliplatin-resistant gastric cancer resistant strain,and the apoptosis rates of SGC-7901 and SGC-7901/L-OHP cell lines at different oxaliplatin concentrations were detected by MTT.2.q RT-PCR was used to detect the relative expression levels of PRRX1 m RNA in both cell lines.3.After transfection of gastric can cer SGC-7901 cell line with interfering PRRX1 lentivirus,SGC-7901 cells differentially expressing PRRX1 were screened and constructed,and the cellular inhibition rate of oxaliplatin on SGC-7901 cell line differentially expressing PRRX1 was detected by MT T assay to further verify the relationship between PRRX1 expression level and the relationship between oxaliplatin resistance in gastric cancer cells.Results 1.The resistance index(RI)of SGC-7901/L-OHP cells to oxaliplatin drug was 6.69,indicating tha t an oxaliplatin-resistant strain of gastric cancer cells was successfully constructed.2.The results of PCR(q RT-PCR)showed that the expression level of PRRX1 in SGC-7901/L-OHP cell line(5.0016 ± 0.41900)was significantly higher than that in SGC-7901 cell line(1.0324 ± 0.30551)(P < 0.05).3.After interfering with PRRX1 lentivirus transfection,the drug sensitivity of SGC-7901 cell line with low PRRX1 expression to L-OHP was increased compared to SGC-7901 cell line with relatively high PRRX1 expressio n and SGC-7901 cell line with PRRX1 empty vector was elevated.Conclusion PRRX1 expression levels were significantly increased in gastric cancer resistant cells,while gastric cancer cells with low PRRX1 expression showed increased sensitivity to oxalipla tin and reduced drug resistance.
