Objective To study the relationship between IGF2BP3 protein in the clinicopathological characteristics of patients with esophageal squamous cell carcinoma(ESCC);the effect of liposome-mediated IGF2BP3-siRNA on the proliferation,migration and invasion of ECA109 cells.Methods Use tissue chip and immunohistochemistry to verify the expression of IGF2BP3 in122 cases of esophageal squamous cell carcinoma tissues,and analysis of the relationship between the clinical and pathological features of patients;Transfection the ECA109 esophageal squamous cell line by mediating IGF2BP3-siRNA,and verify the transfection result in the cell based on the expression of green fluorescent protein;Western blotting method was used to detect the expression of IGF2BP3 protein;Use the scratch test,CCK-8,Transwell to confirm the ability of migration,proliferation and invasion of esophageal squamous cell carcinoma.Results The protein expression of IGF2BP3 in esophageal squamous cell carcinoma was significantly correlated with distant metastasis,lymph node metastasis,and TNM staging(P<0.05),using liposome to transfect ECA109 cells,the transfection efficiency can reach 83.41%.Western blotting results show that the transfected IGF2BP3-siRNA can significantly reduce the expression of IGF2BP3.The results of the scratch test showed that the cell migration distance(137±8.2μm)of the IGF2BP3-siRNA group was significantly lower than that of the NC group(198.4 ± 25.6 μm)and the CON group(228.6 ± 22.5 μ m)(P < 0.05).Decreased expression of IGF2BP3 protein can significantly reduce the migration ability of esophageal cancer cells;In the CCK-8 experiment,the growth inhibition rate of the experimental group at48 hours after transfection was 46.17 ± 0.01%,which can significantly reduce cell proliferation(P﹤0.05);Transwell experiment results showed that the number of invasive cells in the experimental group(84 ± 10.79)was significantly lower than that in the blank control group and negative control group,and IGF2BP3-siRNA can reduce the invasion rate of ECA109 tumor cells(P﹤0.05).Conclusion Patients with esophageal squamous cell carcinoma may have different clinical outcomes due to changes in the expression of IGF2BP3 during clinical progression.IGF2BP3 protein expression in esophageal squamous cell carcinoma is closely associated with clinicopathological features.Decreased expression of IGF2BP3 can inhibit the proliferation,migration and invasion of ECA109 cells.The specific mechanism remains to be verified by our next experiment.Lay a certain foundation for the clinical application of IGF2BP3 in esophageal squamous cell carcinoma.
