Abstract/Summary:
Purpose: To study the difference of miRNA expression between Micro RNA(miRNA)in peripheral blood of patients with non-arteritic anterior ischemic optic neuropathy,NAION)and normal controls,and to predict the relationship between miRNA and NAION by miRNA analysis of the differential expression of miRNA in patients with non-arteritis anterior ischemic optic neuropathy.Method: 1.Peripheral blood RNA was extracted from 6 patients with NAION and 5normal controls for high-throughput mi RAN sequencing,and the filtered sequences were classified and annotated.The differentially expressed miRNA,was analyzed and analyzed by GO,3.The two miRNA with significant differences were verified by real-time quantitative PCR and their biological functions were predicted.Results: 1.A total of 33 miRNAs differential expressions were obtained by high-throughput sequencing,of which 18 were up-regulated and 15 were down-regulated(| log2(foldchange)| > 1,P < 0.05).2.Through GO,KEGG database analysis and bioinformatics prediction,differentially expressed miRNA participates in a variety of biological processes of NAION,such as intercellular signal transduction,angiogenesis,oxidative stress,apoptosis,inflammation and so on.3.The results of q RT-PCR verification of the two miRNA with significant differential expression were consistent with the results of sequencing.Conclusion: 1.This high-throughput sequencing provided NAION-related miRNA differential expression profiles and screened out the abnormally expressed miRNA in NAION.2.It is predicted that differentially expressed miRNA may be involved in multiple signal pathways in the occurrence and development of NAION.This provides a basis for future research on the etiology and treatment of NAION.
