Objective: The character of vancomycin crosslinked acellular bone mass prepared by our research group was analyzed.Methods: Firstly,vancomycin crosslinked acellular bone mass was prepared according to the experimental method of our group.Then the following experiments were carried out :1.The release of vancomycin on cross-linked acellular bone was detected by bacteriostasis test and vancomycin release test.2.microstructure of the extracellular matrix of osteocytes was observed by Scanning electron microscope,and its chemical elements were analyzed EDS energy spectrum.3.the eluted Van-b ECM bone mass for anti-staphylococcus aureus adhesion experiment.4,the cytotoxicity was tested by the extraction solution of Van-b ECM bone mass extracted by the CCK8 experiment.5.the compatibility between osteoblasts and Van-b ECM bone mass was detected by live/dead staining.Results:In this study,vancomycin modified paracellular bone mass was verified and analyzed by the above experimental method,We found that :(1)because of its porous structure,in the process of chemical crosslinking,it is easy to adsorb a large amount of vancomycin to produce cytotoxicity,is not conducive to osteoblast growth.(2)We cleaned the crosslinked bone by PBS buffer,Bacteriostasis test and vancomycin release test showed that it can remove the adsorbed vancomycin after 4 h,Finally,the extracellular matrix bone mass of vancomycin(Van-b ECM).(3)EDS energy spectrum results indicate that the eluted Van-b ECM remains loaded with vancomycin,indicating that the acellular bone of cross-linked vancomycin is difficult to elute.(4)The anti-staphylococcus aureus adhesion test of the eluted Van-b ECM bone mass showed that the cross-linked Van-b ECM had anti-staphylococcal adhesion ability,is an effective antibacterial material.(5)Vancomycin concentration inhibits osteoblast activity once it exceeds 1 mg/ml;live/dead staining showed that osteoblasts could survive on Van-b ECM.To further test the cytotoxicity of the eluted Van-b ECM bone mass by CCK-8 method,Van-b ECM showed low cytotoxicityConclusions: Vancomycin cross-linked acellular bone mass has low toxicity and good biocompatibility to osteoblasts,and has anti-S.aureus effect.It is a very promising antibacterial material.
