Breast cancer is one of the malignant tumors that threaten women’s health.Nowadays,the prevention and treatment of breast cancer’s progress and metastasis has become the focus of research.Therefore,the functional research on breast cancer cells is particularly important.Studies have shown that the hydroxylase JMJD6 may have a hydroxylation modification effect on the histone tyrosine kinase WSTF,and then negatively regulate its protein level.Therefore,in this project,in breast cancer cells,the specific small interfering RNA of JMJD6 was transfected to detect whether the level of WSTF protein was up-regulated.Since both JMJD6 and WSTF are known to have a regulatory effect on the occurrence and development of breast cancer,the subject used CCK8,Trasnwell and scratch experiments to analyze whether JMJD6 regulates the proliferation and migration of breast cancer cells by affecting the amount of WSTF protein.It is known that WSTF can phosphorylate histone H2A.XY142ph,which in turn affects the DNA damage response and regulates the level of H2A.XS139phmodification related to the DNA damage response.Therefore,the subject analyzed whether JMJD6 can regulate the DNA damage response by affecting the amount of WSTF protein after cell DNA damage.Experimental results show that JMJD6 promotes the proliferation and migration of breast cancer cells by down-regulating the expression of WSTF;after DNA damage,JMJD6 affects the level of H2A.XY142phand the response to DNA damage by regulating the amount of WSTF protein.Mechanism analysis showed that JMJD6 affected the transcriptional regulatory activity of WSTF through hydroxylation modification,thereby regulating downstream tumor proliferation and migration-related gene expression,and promoted the degradation of WSTF via the proteasome pathway through the E3 ubiquitin ligase MDM2.The above results show that:JMJD6 can regulate breast cancer cell activity and DNA damage response through WSTF.This result reveals the function of JMJD6 from a new perspective and in-depth study of its mechanism of regulating tumor cell activity,hoping to provide new clues for the treatment of breast cancer.Figure 23;Table 9;Reference 83...
