Implant dentures have a high chewing efficiency,and the foreign body sensations are not obvious.It is an ideal treatment way for patients with missing teeth.However,patients with systemic diseases such as diabetes and osteoporosis may have risks if they undergo implant surgery,which may affect the implant osseointegration.Osteoporosis patients suffer from reduced bone mass,impaired bone microstructure,reduced bone regenerative capacity,and increased bone fragility,which can affect bone healing and thus affect dental implant osseointegration.With the aging of society,the prevalence of patients with osteoporosis continues to increase,and it is urgent to solve the implant problems of patients with osteoporosis.Lycopene has been reported to prevent bone loss in osteoporosis models.However,the role of lycopene in the osseointegration and fixation of dental and orthopedic implants under osteoporotic condition remains unknown.Therefore,We hypothesized that lycopene could improve implant osseointegration under osteoporotic condition.Consequently,the purpose of this experiment is to explore the effect of lycopene on implant osseointegration and fixation in ovariectomized rats.Provides experimental and theoretical basis for the use of lycopene to promote dental implant osseointegration,fixation and long-term results.Methods: Thirty female SD rats were randomly divided into OVX group,OVX+ Lycopene group,and sham group.Rats in the OVX and OVX+Lycopene groups were excised bilateral ovaries.Rats in the Sham group were excised bilateral equal amounts of fat.Twelve weeks after ovariectomy,titanium implants were inserted bilaterally into the distal femoral,On the second day of implant surgery,all rats began to be gavaged:(1)the OVX group: corn oil gavaged(5 ml/kg·bw),(2)the OVX+lycopene group: lycopene dissolved in corn oil(10mg/ml)gavaged(50 mg/kg·bw),and(3)the rats in the sham group were gavaged with corn oil(5 ml/kg·bw).Twelve weeks after gavaged,the rats were sacrificed and both femurs were harvested.For each rat,left femur was used for histological analysis,and right femur was used for biomechanical testing and micro-CT analysis.Results: After 12 weeks of ovariectomy,the total body bone mineral density of the rats showed: OVX group(0.38+0.02)g/cm2,significantly less than Sham group(0.43+0.04)g/cm2(P<0.01),indicating successful osteoporosis model establishment.X-ray shows the high-density shadow of the implants in the three groups of femurs.The OVX group showed a low density between the implants and the bone tissue.The bone density between the OVX+Lycopene group and the bone tissue was similar to that of the surrounding bone,no significant difference with the Sham group.In the OVX group,OVX+Lycopene group and Sham group,BA was(33.18+9.66%),(57.17+8.05%),and(60.99+11.64%),respectively.BA in the ROI of the OVX+Lycopene and Sham groups was significantly higher than that of OVX group,BA of the lycopene group increased by 72.3% compared with the OVX group(P<0.01).BC was(46.53+3.96%),(70.43+10.55%),(73.84+10.93%)respectively.BC in OVX+Lycopene and Sham groups was significantly higher than that in OVX.Compared with OVX group,lycopene group The bone contact(BC)increased by 51.4%(P < 0.01).The maximum pushing power was(33.19+13.23)N,(56.18+20.58)N,and(67.50+12.63)N.The maximum push-out force of OVX+Lycopene and Sham groups was higher than that of OVX group,the maximum push-out power of the lycopene group was 69.3% higher than that of the OVX group(P <0.01),and there was no significant difference of the maximum push-out force between the OVX+Lycopene and the Sham group(p>0.05).BV / TV was(16.12 ± 2.52%,23.51 ± 4.93%,24.09 ± 7.07%),Tb.Th was(100.51 ± 21.60,132.13 ± 11.78,137.59 ± 11.38)μm,and BMD was(141.58 ± 13.56),169.91 ±(594.66±36.88,550.01±35.47,510.30±66.75)μm,Tb.N were(1.49±0.05,1.54±0.13,1.57±0.05)1 /mm respectively.The values of Tb.Th,BMD,BV/TV,and Tb.N in Sham group were higher than those in OVX group(P<0.05),Tb.Sp was lower than OVX group(P<0.05).The values of Tb.Th,BMD,BV/TV in OVX+Lycopene group were higher than those in OVX group(P<0.01),Tb.Sp was lower than OVX group(P<0.05),and the values of OVX+Lycopene and Sham groups were both no significant difference(P>0.05).The micro-CT data of the lycopene group showed that the bone mass and trabecular thickness of the femur were significantly higher than those of the OVX group,and the trabecular space was significantly lower than that of the OVX group.Rats in the OVX group had more bone loss than those in the OVX+Lycopene group and the Sham group,peri-implant bone formation in osteoporotic rats was improved after lycopene treatment.ConclusionsLycopene improved implant osseointegration,fixation and bone formation under osteoporotic condition,suggesting lycopene as a promising therapeutic agent to prevent bone loss and to improve implant osseointegration under osteoporotic condition.
