Objective:To detecte the expression of TRPS1 and GATA3 in primary three negative breast cancer(TNBC),TNBC metastasis and non-TNBC(Luminal A,Luminal B,HER-2 overexpressing breast cancer).Methods:According to inclusion criteria,40 cases paraffin tissue blocks of primary TNBC and 20 cases paraffin tissue blocks of TNBC metastatic were collected as the experimental group,60 cases paraffin tissue blocks of Luminal A breast cancer,and 60cases paraffin tissue blocks of Luminal B breast cancer,and 40 cases paraffin tissue blocks of HER-2 overexpressed breast cancer were collected as the control group,which is underwent radical surgery in Huangshan people’s Hospital from January 1,2016 to December 31,2020.Immunohistochemistry(IHC)was used to detect the expression of TRPS1 and GATA3 in these types of lesions.According to the results of normality test,the measurement data of patients are described by means of mean±standard deviation,and the counting data of patients are described by means of number of cases(percentage).The measurement data were compared between groups by t-test,χ~2test for inter group comparison of count data.Kappa test was used to compare the consistency of GATA3 and TRPS1 expression intensity in patients with different molecular typing.All statistical data were analyzed by SPSS26.0 software,and the difference was statistically significant(P<0.05).Results:1.The positive expression rate of GATA3 was 67.5%(27/40)in TNBC,was75.0%(15/20)in TNBC metastasis,was 100%(60/60)in Luminal A breast cancer,was98.3%(59/60)in Luminal B breast cancer,and was 90.0%(36/40)in HER-2overexpressing breast cancer.The positive expression rate of TRPS1 was 92.5%(37/40)in TNBC,was 95.0%(19/20)in TNBC metastasis,was 98.3%(59/60)in Luminal A breast cancer,was 98.3%(59/60)in Luminal B breast cancer,and was 92.5%(37/40)in HER-2 overexpressing breast cancer.When compared in different groups,the GATA3expression intensity of Luminal A breast cancer and Luminal B breast cancer was significantly higher than that of TNBC and TNBC metastasis,and the TRPS1 expression intensity of Luminal A breast cancer was significantly higher than that of TNBC,the difference was statistically significant(P<0.05).When compared in the same group,according to the consistency test results,the expression intensity of GATA3 and TRPS1 in Luminal A breast cancer,Luminal B breast cancer and HER-2 overexpressing breast cancer were consistent.The expression intensity of GATA3 and TRPS1 in TNBC and TNBC metastases were not consistent.The expression intensity of the two groups was significantly higher than that in the two groups.2.The average age of patients with Luminal A breast cancer and Luminal B breast cancer was significantly higher than that of the other three groups,and the average age of patients with TNBC metastasis was significantly lower than that of the other four groups.The difference was statistically significant(P<0.05).3.The age of patients in GATA3 high expression group was higher than that in negative group and medium expression group(P<0.05).4.There was no significant correlation between the expression of TRPS1 and the basic situation of patients.5.The expression of GATA3 was positively correlated with ER,negatively correlated with proliferation index Ki-67,and had no correlation with HER-2.6.The expression of TRPS1 was positively correlated with ER,but not with HER-2and proliferation index Ki-67.Conclusion:TRPS1 combined with GATA3 can be used as a useful marker in the diagnosis of metastatic TNBC. |
