| Objective:Acute kidney injury(AKI)is a serious illness that is easily seen in clinical work.In recent years,with the continuous increase of morbidity and mortality,the economic pressure of patients is also increasing,which has been paid more attention to it[1].AKI research has been the focus and difficulty of clinical and scientific research.The treatment of AKI is very limited,and there is no drugs have been found to be particularly effective in preventing the further development of AKI.Therefore,it is of great scientific significance and clinical transformation value to actively explore new safe and effective treatment methods for AKI and reveal its mechanism of action.In this study,6-8w male SD rats were selected to prepare a renal ischemia-reperfusion model,and the rats were given resveratrol intragastric treatment to compare Scr,Bun and renal pathological changes of AKI rats in different groups,To study the effect and machine-processed of RES on AKI rats.Methods:SD rats were divided into Sham group、Model group and RES group.All SD rats were adaptively fed for 1 week.RES group:SD rats were given RES(100mg/kg)intragastric treatment for 1 week.After abdominal anesthesia,the kidneys were found and the left and right renal arteries were clamped for 45 minutes,respectively,and then reset.Model group was given the same amount of deionized water intragastric treatment for 1 week,others same as RES group.Sham group:the same amount of deionized water was gavaged for 1week,and the renal artery was opened only without clipping.The levels of Scr、Bun of rats in each group were detected.The molecular protein expressions of PI3K,Akt and m TOR were detected by western blotting.The renal pathological changes were observed by periodate schiff staining.Results:Compared with the Model group,the levels of Scr and Bun were higher than Sham group,the difference was statistically significant(P<0.05).The Scr and Bun of RES group were lower than Model group,the difference was statistically significant(P<0.05).Il-6 was decreased in the RES group compared with the Model group,and the difference was statistically significant(P<0.05).Compared with Model group,the renal tubular epithelial cell oedema,necrosis,inflammatory cell infiltration and other pathological changes were reduced in RES group,suggesting that the renal injury was relatively small.Compared the Model group,it is not significant difference in PI3K、Akt、m TOR and in RES group(P>0.05),p-PI3K、p-Akt and p-m TOR’s expression levels were decreased,the difference was statistically significant(P<0.05).Conclusion:RES can improve the levels of Scr and Bun in AKI rats,inhibit inflammatory response,and alleviate kidney injury.The mechanism may be through the inhibition of PI3K/Akt/m TOR pathway to activate autophagy,and then play a protective role in the kidney. |
