| BackgroundAlcoholic liver disease(ALD)has become one of the serious problems affecting human health all over the world with increasing morbidity and mortality.Developing related functional foods to intervene and protect ALD under the premise of reducing side effects is a research hotspot.Wheat Embryo Globulin(WEG)is a kind of high-quality active plant protein resource,which has anti-inflammatory,anti-oxidation and immunity enhancing activities.In the earlier stage,our research group analyzed KEGG metabolic pathway of WEG based on proteomics,and found that WEG might be involved in metabolic pathway of Alcoholism.On this basis,we studied the protective effect of WEG on alcoholic liver injury in mice.ObjectiveTo investigate the protective effect of WEG in acute and chronic alcoholic liver damage mice.MethodsPreparing WEG by salt extraction,characterized its structure and composition by free amino acid analysis,circular dichroism spectrum analysis and scanning electron microscopy.Male KM mice were selected to establish acute and chronic alcoholic liver injury mouse models by single and continuous gavage of alcohol,respectively.Investigate the effects of WEG on the basic physiological,biochemical indexes and the expression of alcohol metabolism-related proteins at the transcription level of ALD mice.At the same time,the changes of intestinal microflora in ALD mice were analyzed by high-throughput sequencing to further explore the protective mechanism of WEG against alcoholic liver injury.Results(1)Preparation and characterization of WEG: The secondary structure of WEG includedα-helix,β-fold,β-corner and random coiled structure,and the content of random coiled structure was the most.The microstructure of WEG samples showed a large number of irregular flaky structures,rough surface,and some holes.The results showed that WEG had high nutritional value and antioxidant activity.(2)Protective effects of wheat germ globulin on acute alcoholic liver injury in mice: The results showed that compared with the model group,the liver index,spleen index,kidney index,serum total cholesterol(TC),serum triglyceride(TG)and liver malondialdehyde(MDA)contents,and the mRNA relative expression of CYP2E1 in liver of mice in the WEG intervention group were significantly decreased.At the same time,the brain index,liver glutathione(GSH)and superoxide dismutase(SOD)levels were significantly increased.The protection effect of WEG medium dose group was the best.At the same time,WEG has a positive effect on intestinal health to some extent.(3)Protective effects of wheat germ globulin on chronic alcoholic liver injury in mice:The organ index,blood lipid level,liver antioxidant capacity and liver cell apoptosis of mice treated with WEG were all improved to different degrees,and the effect of WEG medium dose group was the best.Further analysis of the intestinal health of mice showed that WEG significantly increased the abundance of intestinal microflora in ALD mice,and the microbial composition also changed greatly.ConclusionsWEG can interfere alcohol-mediated acute and chronic liver injury by improving dyslipidemia,alleviating liver oxidative stress,and regulating mRNA expression of genes related to alcohol metabolism.Based on the enterohepatic axis theory,WEG also plays a certain role in the regulation of intestinal health,further revealing the protective mechanism of WEG on alcoholic liver injury. |
