Research Objective:Neuropathic pain NP is pain caused by damage and lesion of human sensory system due to a variety of physiological and pathological factors.Due to the complex physiological and pathological mechanism which brings great challenges to the clinical management and treatment,the current clinical drugs for the treatment of NP have failed to achieve good results.This disease falls into the category of "blood impediment" in traditional Chinese medicine(TCM)," Different is pain,not glory is pain ".Through the research on Chinese classical prescriptions,we have found that compound Shentong Zhuyu Decoction has good clinical effect on the treatment of congestion swelling and pain,qi stagnation and blood stasis,rheumatism and other conveniences,and it is one of the representative prescriptions for the treatment of impediment,especially blood impediment.The purpose of this study was to observe the effect of Shentong Zhuyu Decoction on NP rat model and its effect on pain behavior of rats,so as to provide effective laboratory evidence for the treatment of NP with Shentong Zhuyu Decoction in clinic.Research methods:Thirty-six male SD rats were randomly divided into a normal group,a normal group(solvent intragastrically),a NP model group(+solvent),a NP model group(+Chinese medicine intervention),a sham operation group(+solvent)and a sham operation group(+Chinese medicine intervention).Three days after modeling,the same experimenter began to gavage the rats in each experimental group.At the same time,the experimental indexes of rats were measured and recorded on the 3rd,5th,7th and 14th days after modeling,such as the mechanical pain threshold value of rats,the thermal pain threshold value and the evaluation and score of locomotor function in rats.On the 14th day after the experiment,the dorsal root ganglia of L4-L6 were selected for further study.Research results:Effects of mechanical pain threshold:from the 3rd day of the experiment,the mechanical pain thresholds of the model group and the sham operation group were decreased to different degrees,and the mechanical pain threshold of the model group was decreased significantly,with a statistical significance(P<0.05).On the 5th and 7th days of the experiment,the mechanical pain threshold of the sham operation group was slightly changed.Compared within the group,the difference between the sham operation group(+Chinese medicine solvent)and the sham operation group(+solvent)was not statistically significant(P>0.05),but within 14 days of the experiment,The results showed that compared with the sham operation group(+solvent),the mechanical pain threshold of the sham operation group(+solvent)was slightly increased and the difference was statistically significant(P<0.05).The mechanical pain threshold of the model group was observed.On the 5th,7th and 14th day of the experiment,the mechanical pain threshold of the model group(+Chinese medicine solvent)was increased significantly,while that of the model group(+solvent group)was not changed significantly,and the difference was statistically significant(P<0.05).Effects of thermal pain threshold:From the third day of the experiment,the thermal pain thresholds of the model group and the sham operation group were decreased to different degrees.Compared with the model group and the sham operation group,the thermal pain threshold of the model group was decreased more significantly,and the difference was statistically significant(P<0.05).On the 5th,7th and 14th day of the experiment,the thermal pain threshold of the sham operation group was slightly increased.Compared with the sham operation group(+solvent)on the 14th day,the thermal pain threshold of the sham operation group(+solvent)was increased significantly(P>0.05).On the 5th,7th and 14th day of the experiment,the thermal pain threshold of the model group(+Chinese medicine solvent)was increased significantly,and the difference was statistically significant as compared with the model group(+solvent)(P<0.05).Effects of behavioral function on left hind limb:On the third day of the experiment,rats in the model group and the sham operation group showed different degrees of motor dysfunction,and the motor dysfunction of the model group was more significant.Compared with the sham operation group,the difference was statistically significant(P<0.05).On the seventh day of the experiment,there was no obvious abnormality in the locomotor function of the sham operation group.Compared with the model group(+solvent)on the seventh day and the fourteenth day of the experiment,the locomotor function of the model group(+solvent)was significantly improved in rats with the model group(+solvent)and the difference was statistically significant(P<0.05).Conclusion:Through the above experiments,we have found that Shentong Zhuyu Decoction can effectively inhibit the mechanical pain and thermal hyperalgesia in SNL model rats,and Shentong Zhuyu Decoction can improve the motor function of SNL model rats,thus promoting the relief of pain symptoms and the recovery of motor function.
