| Objective:In this study,the BDNF serum level and gene polymorphism of autistic children and the control group were studied respectively and we compared the different distributions of alleles and genotype frequencies in children with autism in single-parent and double-parent families,and thus to explore the influencing factors of childhood autism.This exploration can provide some reference for the study of problems associated with BDNF gene(including childhood autism),and is expected to provide new clues for clarifying the etiology and pathogenesis of childhood autism,and contribute to the prevention and treatment of autism.Methods:In this study,a case-control method was adopted.The subjects were children with autism who were admitted to the Department of Psychiatry,Hainan Hospital Affiliated to Hainan Medical University and the Department of Children’s Rehabilitation,Anning Hospital,Hainan Province from September 2019 to December2020.According to the inclusion and exclusion criteria of the subjects,78 children in the case group were determined,80 control children who had undergone physical examination in pediatrics or health care departments of hospitals in corresponding periods were selected at the same time.Their baseline data were collected,the Autism Rating Scale for Children was evaluated,and the scores of Eysenck Personality Questionnaire(adult version)were measured.The 5-10 m L blood from anterior elbow vein of autistic children and healthy control children were drawn for serum BDNF level determination and gene polymorphism analysis.Results:1.The serum level of BDNF in autistic children was higher than that in healthy control children[(19.89±6.92vs13.34±3.82)ng/ml](t=7.390,P<0.05).2.There are gene polymorphisms at three SNPs loci,rs6265,rs10835210 and rs11030104 in the population of Hainan.The allele and genotype frequencies conform to the Hardy-Weinbery genetic balance test(P>0.05).3.We compared the effects of three SNPs rs6265,rs10835210 and rs11030104 on BDNF serum levels in autistic children.The results showed that there were significant differences in the three genotypes AA,AG and GG of BDNF rs6256 and BDNF rs11030104 in the experimental group(F=3.759,P=0.028)(F=3.763,P=0.027).In the comparison of GG genotype with AA and AG genotypes of the two loci,the BDNFserum level of patients with GG genotype was the highest[(19.31±5.46)ng/ml]and[(19.46±5.58)ng/ml].There was no significant difference in the comparison of AA,AC and CC genotypes of BDNF rs10835210locus(F=2.105,P=0.128).4.Correlation analysis between CARS score and BDNF serum level of autistic children from single-parent or recombined families and double-parent families showed that the CARS score of children from single-parent families or recombined families was higher than that of children from double-parent families,and the difference was statistically significant(t=2.933,P=0.004),and there was no difference in BDNF serum level(t=-0.268,P>0.05).The three SNP loci rs6265,rs10835210 and rs11030104 in the BDNF gene were compared according to single-parent families and recombinant families and double-parent families.The comparison results showed that there was no difference in the three SNP gene loci.Stratified analysis of rs6265 genotype,rs10835210 genotype and allele distribution according to CARS score showed that there were significant differences between mild-moderate and severe patients[(χ~2=13.067,P=0.001),(χ~2=6.942,P=0.031)and(χ~2=6.103,P=0.013)].5.Compared with the parents of children in the healthy control group,the parents of autistic children have differences in the measurement of four dimensions of Eysenck Personality Questionnaire(P<0.05).The scores of introversion-extroversion(E dimension)and concealment(L dimension)of parents of autistic children were lower than those of parents of children in the healthy control group,while the scores of psychoticism(N dimension)and emotional stability(P dimension)of parents of autistic children were higher than those of parents of children in the healthy control group,and E and P dimensions were independent risk factors for autism[P=0.020,OR(95%CI)0.945(0.901~0.991)]and[P=0.010,OR(95%CI)1.073(1.017~1.133)].Conclusion:By measuring the serum level of BDNF and its genetic polymorphism and referring to the general scales at home and abroad to evaluate the influencing factors of autistic children,this study found that the rs6265,rs10835210 and rs11030104 loci in BDNF gene were polymorphic in children in Hainan area,and the genotypes of rs6265,rs10835210 and their alleles were different among patients with different severity of diseases,suggesting that the above two loci may be susceptible genes affecting the severity of autism.Secondly,the high expression level of serum BDNF,single parent family or recombined family or parental personality defects may have influence on the incidence and severity of autism spectrum disorder children. |
