| Objective:To observe the effect of endogenous phlegm dampness on serum adipofactor Vaspin in CIA mice,in order to understand the important role of endogenous phlegm dampness in RA more comprehensively.Materials and Methods:In total 32 SPF 4-week-old DBA/1 male mice were randomly assigned based on the random number table method: blank control group(BC group,n=8),phlegm-dampness control group(PD group,n=8),non-phlegm-dampness CIA group(NPDC group,n=8)and phlegm-dampness CIA group(PDC group,n=8).Among them,the BC group and NPDC group were fed with common feed,PD group and PDC group were fed with high-fat.After 28 days of feeding,the CIA models were made in NPDC group and PDC group,while the BC group and PD group were injected with normal saline.On day 70,the mice were sacrificed.During the experiment,the body weight of mice was measured and the changes of arthritis in mice were observed.After the mice were taken,HE staining was used to evaluate and analyze the histomorphological changes of the ankle joint.Serum lipid,IL-6 and adipocytokine Vaspin levels were detected by ELisa.Results:1.Phlegm-dampness of mice in each group: From day 1 to 28,mice in BC group and NPDC group had good spirit,color,shape and state,and normal excreta color.The mice in PD group and PDC group were lazy,lethargic,unresponsive,obese,dull and dull,testicular sagging,low food intake and yellow feces.On the 29 th and70th day after CIA modeling,there was no significant change in BC group and PD group.The NPDC group mice began to tremble,bow back,like to get together and fear the cold,lose weight and lose hair,but there was no obvious abnormality in feces.The mice in PDC group had severe trembling,mental malaise,dull squinting eyes,unpleasant movement,curled back of bow,haggard fur,red anus,drooping testicles,yellow feces and sparse water.2.Body weight changes of mice in each group: 28 days ago,the body weight of mice in each group showed an increasing trend.After the 28 th day(mouse modeling),the weight of mice in PDC group began to decline,and the weight of mice in the other three groups continued to increase.After 49 days(modeling 21 days),the weight of mice in PDC group and NPDC group showed a decreasing trend,especially in PDC group.3.Joint AI score of mice in each group: The AI score of mice in BC group and PD group was 0.On the 21 st day,the AI score of mice in NPDC group began to increase.AI scores of mice in the NPDC group and the PDC group increased continuously at 21-33 days.and the change of AI score in PDC group was the most obvious.From day 33 to day 42,the AI index of mice in NPDC group and PDC group continued to increase,and the increase of AI index was slow.On the 42 nd day,there was significant difference in AI score between NPDC group and PDC group(P<0.01).4.Histomorphological observation results of knee joints in each group: Morphological results of mice indicated that there was no abnormal tissue structure of knee joint in BC group.In the PD group,no other arthritis-related pathological changes were observed except a small amount of inflammatory cell infiltration.Compared with the PD group,the articular surface of mice in the PDC group was slightly rough with more inflammatory cell infiltration.Compared with the NPDC group,the articular surface of the mice in the PDC group was rough,the synovial tissue was hyperplasia,a large number of inflammatory cells infiltrated,and the articular space was full of pus.5.Blood lipid of mice in each group:Compared with the BC group,the levels of TG,TC and LDL-C in the PD group and the PDC group were significantly increased(P < 0.01),and the level of HDL-C was significantly decreased(P< 0.01).The TG and TC in the NPDC group had an increasing trend,but there was no statistical significance(P>0.05).LDL-C was significantly increased and HDL-C level was significantly decreased(P<0.01).Compared with the PD group,the TC and TG levels in NPDC group were significantly decreased(P<0.01).Compared with NPDC group,TG and TC of mice in PDC group were significantly increased(P<0.01).6.Serum IL-6 and Vaspin levels:Compared with BC group,serum IL-6 concentration in the other three groups was significantly increased(P<0.05);Compared with the PDC group,the serum IL-6concentration in the PD group and the NPDC group was significantly decreased(P<0.05).Compared with BC group,serum Vaspin concentration in the other three groups was significantly increased(P<0.05);Compared with the PDC group,the concentration of Vaspin in the PD group and the NPDC group was significantly decreased(P<0.05).7.There was a positive correlation between Vaspin and TC,the correlation coefficient r=0.272,P=0.000.There was a positive correlation between Vaspin and IL-6,with correlation coefficient R =0.561,P=0.000.Conclusion:1.Phlegm-dampness factor caused high AI index in CIA mice.2.Phlegm-dampness factor aggravated the pathological changes of joints in CIA mice.3.Phlegm-dampness factor aggravate the degree of arthritis by up-regulating Vaspin and IL-6 in serum of CIA model mice. |
