The Effect Of LncRNA-ATB On Proliferation,migration And Apoptosis Of Breast Cancer And Its Clinical Significance | | Posted on:2022-01-12 | Degree:Master | Type:Thesis | | Country:China | Candidate:A N Liu | Full Text:PDF | | GTID:2504306731452414 | Subject:Clinical Laboratory Science | | Abstract/Summary: | PDF Full Text Request | | Objective: This research was designed to investigate the effect of lncRNA-ATB on the proliferation,migration and apoptosis of breast cancer MCF-7cell and MDA-MB-231 cell,and to investigate the expression and clinical significance of lncRNA-ATB in serum of patients with breast cancer.Methods: The cultured breast cancer cell MCF-7 and MDA-MB-231 were divided into 5 groups: blank group(no treatment),si-NC group(transfected negative sequence),si-ATB(transfected specific interferingRNA,designed and synthesized 3 specific siRNA interference sequences,a total of 3 groups)respectively.The expression of lncRNA-ATB was detected by real-time PCR to verify the transfection efficiency and screen the most suitable interference sequence for subsequent experiments.The ability of cell proliferation was tested by colony formation assay,the migratory ability was evaluated by transwell migration assay and the cell apoptosis rate was examined by flow cytometry.The serums of 50 patients with breast cancer,30 patients with benign breast tumor and 34 healthy subjects were selected from The Second Affiliated Hospital of Hunan Normal University during February 2019 to September 2020.The expression levels of lncRNA-ATB and CA153 in serum was detected and analyzed expression differences of the two indexes among the three groups.The ROC curve of the two indexes in the diagnosis of breast cancer was drawed to evaluate the diagnostic value for breast cancer.The clinical data of breast cancer patients were collected to analyze the relationship between the expression level of lncRNA-ATB and clinical data.Results: 1.In the two breast cancer cells MCF-7 and MDA-MB-231,the level of lncRNA-ATB in the blank group and the si-NC group are significantly higher than that in the si-ATB-1,2,and 3 groups(P<0.05).The expression level of si-ATB-1 is the lowest in these groups,and it was selected as the most suitable interference group for subsequent experiments.Compared with the blank group and si-NC group,the abilities of colony formation and migratory in si-ATB group were significantly reduced(P<0.05),while the cell apoptosis rate of the siATB group was significantly increased(P<0.05).2.The expression level of lncRNA-ATB in serum of patients with breast cancer was significantly higher than that in benign breast tumor group and the healthy subjects group(P<0.05);the area under ROC curve of lncRNA-ATB was significantly higher than that of CA153(P<0.05);the expression level of lncRNA-ATB in patients with lymph node metastasis was higher than that of patients without lymph node metastasis(P<0.05),and the expression level of lncRNA-ATB in patients with advanced breast cancer was higher than that of early breast cancer(P<0.05);the level of lncRNA-ATB in patients with high Ki67 expression was higher than that in patients with low Ki67 expression(P<0.05).Conclusion: 1.Down-regulating the expression of lncRNA-ATB can inhibit breast cancer cell proliferation and migration,and promote cancer cell apoptosis;2.The expression level of lncRNA-ATB in the diagnosis of breast cancer is better than CA153,and its expression level is related to the level of lymph node metastasis,TNM stage and Ki67 expression in patients.High expression of lncRNA-ATB may indicate poor prognosis.It can be considered as a new type of serum marker for breast cancer in clinical auxiliary diagnosis and prognosis evaluation. | | Keywords/Search Tags: | lncRNA-ATB, breast cancer, proliferation, migration, apoptosis, marker | PDF Full Text Request | Related items |
