Objective: The aim of this study was to observe the effects of acupoint stimulation(AS)on the intestinal barrier function in hepatic ischemia-reperfusion injury(HIRI)rats through animal experiments,and to explore the preoperative Electric acupuncture(EA)can stimulate the Dachangshu(BL25)and Zusanli(ST36)points to reduce the intestinal injury of HIRI rats and whether it has protective effects on the intestinal function in hepatic ischemia-reperfusion(I/R)rats,and provide a basis for the application of AS in the clinical perioperative period.Methods:32 male SD rats(180~210 g)were divided into Sham group(Sham),ischemia-reperfusion group(I/R),acupoint-stimulated+I/R(AS+I/R)and non-acupoint-stimulated+I/R group(n AS+I/R),each group were 8.After 1 h of ischemia and 4 h of reperfusion,inferior vena cava blood specimens and small intestine samples of rats were taken respectively,Lai’s methods were used to detect alanine aminotransferase(ALT)and aspartate aminotransferase(AST)concentrations,Chiu’s evaluation method was used to evaluate the degree of intestinal mucosal damage of small intestine tissue under microscope after HE staining,ELISA method was used to detect serum diamine oxidase(DAO)and DLactic acid(D-Lac),immunohistochemistry(IHC)was used to detect the expression of ZO-1 tight junction(ZO-1)protein in small intestine.Results:(1)Serum AST and ALT levels in I/R group were significantly higher than those in Sham group,AS+I/R group and n AS +I/R group(P<0.05).(2)The Chiu’s score of small intestine tissue in the I/R group was higher than that in the Sham group,while in the AS+I/R group was lower than that in the I/R group and the n AS+I/R group,but the degree of decrease in the n AS+I/R group was not as good as that in the AS+I/R group(P<0.05).(3)Serum DAO and D-LAC in I/R group were higher than those in Sham group,and intestinal barrier function was seriously impaired.Serum DAO and D-LAC in AS+I/R group were lower than those in I/R and n AS+I/R groups,but the degree of decrease in n AS+I/R group was not as good as that in AS+I/R group(P<0.05).(4)Compared with Sham group,ZO-1 protein expression was lower in I/R group,and intestinal mucosal tissue integrity was decreased,while ZO-1 protein expression was increased in AS+I/R group compared with I/R group and n AS +I/R group,but the degree of increase in the n AS +I/R group was not as good as that in the AS+I/R group(P<0.05).Conclusion: Preoperative EA on Dachangshu and Zusanli points can alleviate the intestinal damage of HIRI rats,and protect the intestinal barrier function.Electroacupuncture stimulation of non-acupoint parts also plays a role.
