ObjectiveThe relationship between intestinal flora and human body is complex,which runs through a variety of physiological activities and pathological processes.Under normal circumstances,maintain a dynamic balance.Long-term use of mixed antibiotics can lead to intestinal flora imbalance.The imbalance of intestinal flora will not only affect the functional state of the intestinal tract,but also have an extensive impact on the immune status and function of the body.Whether the imbalance of intestinal flora caused by antibiotics will make the influenza virus infection in lung tissue more serious,and whether there is a specific relationship between intestinal flora and TLR7 signaling pathway it is not clear.The aim of this study was to establish an influenza mouse model of intestinal flora imbalance induced by antibiotics,and then infect the lung adaptive strain of influenza A virus FM1,and then explore the effect of intestinal flora imbalance on TLR7/NF-κB signal pathway in the lungs of influenza mice through probiotics,FMT,Guizhi Mahuang Gebantang and Shenling Baizhu San.MethodIn the study of probiotics and fecal transplantation,C57BL/6J mice were divided into normal control group(Blank),virus control group(Virus),antibiotic treatment+virus group(Model),model+probiotic recovery group(Probiotics),model+fecal microbiota transplantation group(FMT).In the study of compound traditional Chinese medicine,the TLR7 knockout mice of C57BL/6J mice were divided into normal control group(Normal),virus control group(Virus),antibiotic treatment+virus group(Model),model+Guizhi mahuang decoction(GZMHGBT),model+Shenling Baizhu powder(SLBZS).Then the changes of 16 sr RNA gene sequencing,body weight,lung index,plaque test,histopathology,T cell subsets and TLR7/NF-κ B signal pathway were detected.ResultIn the model of intestinal flora imbalance in mice,Enterobacteriaceae accounted for40.4%-84.0%,and Tannerellaceae also significantly increased(35.4%-54.6%),while other families of Bacteroides(39.2%-52.8%),Spirillaceae(10.8%-21.6%)and Provodiaceae(8.00%-9.70%)in normal intestinal flora were seriously suppressed.the number,richness and diversity of intestinal flora decreased.The use of compound traditional Chinese medicine,probiotics and fecal transplantation can effectively restore the destroyed intestinal flora,alleviate the degree of weight loss in mice,and reduce the inflammatory damage of lung and cecal tissue.Decrease the overexpression of TLR7/NF-κB signal pathway and improve the balance of TH cells.ConclusionTraditional Chinese medicine,probiotics and fecal microbiota transplantation can restore the richness,diversity and composition of intestinal flora in influenza mice,improve the balance of TH cells,reduce the inflammatory reaction of lung tissue and cecum,and play an antiviral role by down-regulating TLR7/NF-κ B signal pathway of immune cells in the lung.
