| Objective: Colorectal cancer is a common malignant tumor of digestive tract.Dysbacteriosis of intestinal flora and circadian rhythm disorder not only participate in the occurrence and development of colorectal cancer,but also play an important role in chemotherapy resistance of colorectal cancer.It has also been reported that not only the circadian rhythm of the host affects the composition and function of the intestinal flora,but also the intestinal flora is involved in regulating the circadian rhythm of the host.The aim of this study was to investigate the effect of pathogenic Escherichia coli on chemoresistance of colorectal cancer by regulating the expression of circadian protein Per1.Methods:(1)Western blot analysis showed that pathogenic Escherichia coli and its metabolite,small molecular compound ARN-5187 influence the expression of circadian rhythm PER1 protein;(2)MTS analysis shows that the effect of pathogenic Escherichia coli and its metabolites on the growth of colorectal cancer cells,the effect of pathogenic Escherichia coli and its metabolites on the growth of colorectal cancer cells by regulating the expression of circadian rhythm PER1 protein,and the effect of pathogenic Escherichia coli and its metabolites on the chemoresistance of colorectal cancer cells by regulating the expression of circadian rhythm PER1 protein;Results:(1)Western blot showed that pathogenic Escherichia coli and its metabolites inhibited the expression of circadian rhythm Per1 protein,and small molecular compound ARN-5187 increased the expression of circadian rhythm Per1 protein;(2)MTS analysis shows that the growth of colorectal cancer cells was stimulated by pathogenic Escherichia coli and its metabolites by inhibiting the expression of circadian rhythm PER1 protein,the chemoresistance of colorectal cancer cells was increased by pathogenic Escherichia coli and its metabolites by inhibiting the expression of circadian rhythm PER1 protein.Conclusion: Small molecular compound ARN-5187 can increase the expression of circadian rhythm PER1 protein and inhibit the growth of colorectal cancer cells by inhibiting REV-ERB pathway;pathogenic Escherichia coli and metabolites can promote the growth of colorectal cancer cells by inhibiting the expression of circadian rhythm PER1 protein;pathogenic Escherichia coli and its metabolites increase chemoresistance of colorectal cancer cells by inhibiting the expression of circadian rhythm PER1 protein. |
