Objective:Indole alkaloids are widely present in nature,and most of them have a wi de range of pharmacological activities including anti-tumor,antibacterial and an ti-inflammatory,and are one of the hotspots of scientific research.However,th e output of natural separation is limited and cannot meet the current demand f or medicine.This thesis focuses on the synthesis and research of the indole al kaloid Ellipticine and its derivatives,and develops a new synthetic method to prepare it for the synthesis of the same type of indole alkaloid.Methods:According to the search system Scifinder,it was found that there are man y related literature reports,but the synthesis route is relatively cumbersome,the experimental conditions are demanding,and the yield is not high.In this rese arch method,the simple reagents 3-acetylindole and 3-amino-1-propanol were u sed as starting materials,and a total of 8 steps were carried out to prepare the key fragments of the ring-closure precursor,3-(1H-indole)-3-yl)acrylic acid an d(E)-5-aminopent-2-enoic acid methyl ester hydrochloride,the two fragments a re then coupled to obtain the ring-closure precursor,and then O2 is used as th e terminal oxidant,ethylene glycol Under the condition of the solvent,the reac tion was stirred at 120℃ for 24 h to obtain 4a with a higher yield.Finally,t he total synthesis of ellipticine was achieved through a 5-step reaction such as redox reaction.The core of the synthesis of key step 4a lies in the use of a key series reaction(three reaction processes including CH activation-Diels-Ald er reaction-oxidative dehydrogenation)to simultaneously close the C ring and D ring in the molecular structure of the compound.ring.At the same time,thi s paper is extended with indole with different substituents to verify the applica bility and scalability of the method.Results:The research method of this thesis successfully synthesized Ellipticine,and obtained 16 indole alkaloids with ellipticine Ellipticine as the core.Conclusion:In this thesis,a simple reagent is used as the starting material,and a new synthetic method for constructing the basic skeleton of ellipticine has been successfully developed,and it has been extended in the synthesis of its derivatives.
