| Purpose:We tried to develop a hydrogel with good biocompatibility for loading curcumin.The feasibility of the hydrogel as a curcumin delivery platform and the effect of hydrogel loading curcumnin the repair of peripheral nerve injury were investigated through experiments.Method:(1)Prepare a series of human hair keratin/chitosan hydrogels of different ratios to study characterization of hydrogels,including FTIR,mechanical properties,porosity,pore size,Water uptake capacity,cytotoxicity,and cytocompatibility.Determine the optimal ratio of keratin/chitosan hydrogel preparation by comprehensively analyzing different characterization results(2)Curcumin was added during the preparation of the hydrogel and the effect of the ratio of human keratin and chitosan on curcumin release was studied.(3)Select the appropriate proportion of the compound hydrogel to load curcumin and perform animal experiments.The effect of composite hydrogel on peripheral nerve injury repair was comprehensively analyzed by animal behavior,histology,electrophysiological,and target organ recovery.Result:(1)Through the process of casting,freeze drying and sodium tripolyphosphate crosslinking,keratin/chitosan hydrogel can be prepared,and the mechanical properties,pore size and porosity of the hydrogel can be adjusted by changing the ratio of keratin/chitosan,of which the proportion of chitosan can improve the mechanical properties of the hydrogel.(2)Keratin/chitosan hydrogel has good cytocompatibility and can be used as a carrier to support cell growth,of which the increase in the proportion of keratin can improve hydrogel cytocompatibility.(3)Curcumin can promote the migration of RSC96 cells at a certain concentration Cucumin can be loaded with the hydrogel and released slowly,of which the increase in the proportion of keratin can speed up the release process.(4)The experimental results of mechanical strength,cytocompatibility and curcumin delivery were comprehensively analyzed to determine the optimal proportion of hydrogel preparation.Animal experiments were conducted using a hydrogel of 1:1 for human keratin:chitosan.In animal experiments,complex hydrogels Loaded with curcumin showed positive effects on behavior,histology,electrophysiological and target organ recovery in rats with nerve injury.Conclusion:Human hair keratin/chitosan hydrogel with mechanical strength,good cell and biocompatibitity was prepared by a simple process.Complex hydrogel can be used as curcumin sustained-release carrier.The complex hydrogel loaded with curcumin showed positive effects on the behavior,histology,electrophysiological and the recovery of target organs in injured rats.Human hair keratin/chitosan hydrogel loaded with curcumin can be used as a therapeutic strategy for nerve injury repair,to integrate the advantages of different substances for nerve injury repair. |
