Sanghuang is a rare medicinal fungi mainly distributes in China,Japan,Korea and other Asian regions.As a treasurable medicinal mushroom,Sanghuang was first recorded in the“Shen Nong Materia Medica”as well as other late ancient herbal pharmacopeia,such as“Ben-Cao-Gang-Mu” in China.Sanghuang was recorded to treat the diseases including metrorrhagia,stranguria with blood,anorectal bleeding,leucorrhea,amenorrhea and spleen-deficiency diarrhea,etc.Modern studies have shown that Sanghuang had various pharmacological effects such as anti-cancer,immunomodulation,anti-diabetes,antioxidant and anti-inflammation,etc.The high preventive and therapeutic effect of Sanghuang against cancers in nucleur-radiation areas of Japan attracted more lots of attention and the medicinal and health-promoting values of Sanghuang were accept by the world.Presently,the wild Sanghuang can not meet the demand in market,and cultivative Sanghuang is the main raw material for medicinal usage.Objective: To compare the effects of different varieties and cultivation modes of Sanghuang on immunomodulation in vivo and antitumor activities in vitro.To provide suggestions for the better development of Sanghuang.Methods: 1.The contents of total polysaccharide,total flavonoid,total phenol,total triterpene and ergosterol in five different Sanghuang varieties were evaluated by means of sulfuric acid-anthrone,forlin-phenol,nitrite-aluminum nitrate,oleanolic acid and high performance liquid chromatography.2.A mouse immunosuppression model was constructed with cyclophosphamide.The in vivo immunomodulative effects of the five Sanghuang aqueous extracts on the cyclophosphamide-induced mice were measured by the peripheral immune organ indices,the proliferative transformation ability of splenic lymphocytes induced by Con A and LPS,and the TNF-α and IL-2 contents in serum.3.The in vitro anti-tumour effects of the aqueous extracts of the five Sanghuang were determined in CCK-8 assay.Conclusion: 1.The content of the active ingredients of the five Sanghuang varieties were differeent,which was related to their varieties and cultivation modes.2.The Sanghuang extracts could improve the immune function of mice by increasing the index of immune organs in immunocompromised mice;increasing the serum content of IL-2 and TNF-α,and promoting the proliferation of spleen lymphocytes in mice.In addition,the Sanghuang varieties with higher polysaccharide content had a higher immune-improving effect.3.The extracts of five Sanghuang varieties had different cell proliferation inhibitory effects against five types of human-derived tumor cells.Moreover,the Sanghuang varieties with higher total flavonoid and total phenolic contents had stronge rantitumour activity in vitro.
