| “Sanghuang” was first recorded in Shennong Materia Medica Classic,which is a kind of large fungus medicinal material mainly belonging to genus Sanghuangporus.In recent years,“Sanghuang” have been widely concerned and studied for their remarkable anti-tumor effects.The composition of “Sanghuang” has polysaccharides,flavonoids,terpenoids,steroids,etc.,it has certain effects incliding anti-tumor,anti-oxidation,neuroprotection,anti-virus,anti-inflammation,antioxidation,hypoglycemia,immune regulation and so on.Sanghuangporus vaninii is the most easily cultivated species of Sanghuangporus,as well as the most common variety in the market.In 2019,S.vaninii were formally recorded into the Processing Specifications of Traditional Chinese Medicine Decoction Pieces in Anhui Province and could be clinically used.However,the clinical application of its decoction pieces is still limited.At present,there are few studies on the efficacy of S.vaninii in immune regulation.On the one hand,the immune deficiency model was established in this study to prove its efficacy in immune regulation.On the other hand,a mouse model of liver cancer was established to preliminarily explore the anti-tumor and immune improvement effects of S.vaninii combined with chemotherapy drugs.The aim is to expand the application range of S.vaninii decoction pieces and provide reference for clinical applicationObjective: HPLC fingerprint of S.vaninii decoction pieces was established.On the one hand,to investigate the effect of S.vaninii on immune function of immunocompromised mice induced by cyclophosphamide,on the other hand,to preliminarily explore the antitumor and immunomodulatory effects of S.vaninii combined with fluorouracil on hepatocellular carcinoma H22 mouse model.Methods: In this study,S.vaninii were used as raw materials,the water extract was extracted by ultrasonic and boiling water,and its fingerprint was established by HPLC.Mice were given S.vaninii for 21 days,and cyclophosphamide was injected in the last three days of administration to cause immune dysfunction in mice.The regulating effect of S.vaninii on immune organs was evaluated by measuring spleen and thymus index and HE staining morphology of spleen.The contents of immunoglobulin and cytokines in serum of mice were determined by ELISA to evaluate the effect of S.vaninii on the content of immune molecules.The function of immune cells was evaluated by spleen lymphocyte proliferation,carbon clearance and ear swelling.H22 cells were subcutaneously injected into tumor-bearing mouse models,and tumor volume,tumor weight and spleen weight were recorded in each group.The pathological changes of tumor tissue and spleen were observed by HE staining,and the antitumor and immune effects of S.vaninii combined with fluorouracil were preliminarily discussed.Results: The fingerprint of S.vaninii was established and four components were identified by chemical standard,which were protocatechuic acid,protocatechuic aldehyde,4-(3,4-dihydroxybenzene)-3-butene-2-one and ergosterol.In the model experiment,the modeling method of model Ⅳ group was selected for the subsequent experiment.The results of animal functional experiment showed that the indexes of the model group were significantly lower than those of the normal group,and the model was successful.Compared with model group,spleen and thymus index of mice treated with water extract of S.vaninii increased,and He staining of spleen also indicated that water extract of S.vaninii could improve the pathological state of spleen tissue caused by cyclophosphamide.The levels of immunoglobulin and cytokines in serum of mice also increased significantly.The proliferation degree of Con A/LPS spleen lymphocytes,phagocytic rate and phagocytic index of macrophages and ear swelling degree of mice were increased.In vivo experiments of hepatocellular carcinoma mice showed that compared with the model group,the average tumor weight and relative tumor volume of fluorouracil group,S.vaninii group and combined administration group were significantly decreased,and HE staining of tumor tissue showed that the morphological structure of tumor cells in each administration group was destroyed.Compared with the fluorouracil group,the spleen index of mice in the combined administration group was increased,and HE staining of spleen showed that S.vaninii could improve the pathological changes of spleen tissue in tumor bearing mice undergoing chemotherapy.Conclusion: The study showed that the water extract of S.vaninii had stable quality,had good immune regulation effect on immunosuppressive mice induced by cyclophosphamide,and had good improvement effect on immune organ injury,immune cell function reduction and cytokine secretion reduction.S.vaninii can significantly inhibit the tumor growth of H22 hepatocellular carcinoma mice,and enhance the immune function and synergistically enhance the effect of chemotherapy in mice to a certain extent. |
