Abstract/Summary:
Objective: In this study,angiotensin Ⅱ-induced AAA mouse model was established to observe the changes of MMP-2/MMP-9,CD147,Cy PA expression,apoptosis and autophagy,and to study the vascular smooth muscle cells in the process of Ang Ⅱ-induced aortic aneurysm formation.The role of autophagy.Methods: The35-48-day-old SPF adult male healthy APOE-/-mice were adaptively reared to 10 weeks of age and then divided into the control group(group A): APOE group;experimental group(group B): APOE induced by Ang Ⅱ Groups;6 in each group,12 in total.Group B was implanted with Angll sustained-release pump [1,000 ng/min/kg]subcutaneously to establish animal AAA model,and group A was implanted with the same dose of normal saline pump subcutaneously.After 28 days,the abdominal aorta was harvested and preserved in 4% paraformaldehyde and 4% glutaraldehyde.α-SMA,Beclin-1 and Bcl-2 were detected by immunofluorescence to detect the autophagy and apoptosis of vascular smooth muscle in plaques,and the levels of MMP-2/MMP-9 were detected by QPCR;the protein expressions of CD147 and Cy PA were detected by WB,and the observation by transmission electron microscopy Autophagy in vascular plaques.Results: The formation of abdominal aortic aneurysm in APOE-deficient mice mediated by Ang Ⅱ was observed by naked eyes,and the model was successfully established.Compared with the control group,the expression level of Beclin-1 in the abdominal aortic aneurysm model of apo E-deficient mice induced by Angll was reduced,and Bcl The expression level of-2 increased,and decreased autophagosome formation in vascular plaques,the m RNA levels of aortic MMP-2 and MMP-9increased,and the protein expressions of CD147 and Cy PA increased.Conclusion:Vascular smooth muscle autophagy decreased and apoptosis increased during Ang Ⅱ-induced AAA formation.Angll-induced VSMC apoptosis and autophagy inhibition in apo E-deficient mice can lead to the formation of abdominal aortic aneurysm.The increased levels of MMP-2/MMP-9 m RNA and the high expression of Cy PA and CD147 may be related to the formation of abdominal aortic aneurysm caused by autophagy inhibition in vascular smooth muscle cells.
