At present,the dietary structure of Chinese people is diversified,the intake of highpurine food is increasing,and the incidence of hyperuricemia and other diseases is also increasing significantly.Although attention has been paid to hyperuricemia and gout,the comprehensive management of uric acid in patients with gout is still not ideal.The side effects of current drug treatment should not be ignored and strict diet control is extremely difficult.It has been reported that intestinal flora has a certain relationship with the incidence of hyperuricemia.As beneficial bacteria in the human intestine,lactic acid bacteria also have beneficial nature.Oral lactic acid bacteria may be used as a new method to treat hyperuricemia and gout by interfering uric acid metabolism.Therefore,this study screened the lactic acid bacteria with high efficiency in absorbing and degrading purine nucleosides,and explored its physiological and probiotic characteristics.By constructing high uric acid hematic disease rat model to evaluate the strain of high uric acid hematic disease,and the ease of gouty nephropathy caused by therapeutic effect,and is based on analysis of the transcriptome study against the strain specificity of purine metabolic pathways in the related research,so as to micro ecological treatment of high blood uric acid and gout disease development provides new ideas.The main research results are summarized as follows:1.A strain of Lactobacillus brevis with strong ability of absorbing and degrading purine nucleoside was screened out,which was identified as Lactobacillus brevis PDD-5,and the storage number was CGMCC No.20573.The absorption rates of inosine and guanosine were 68.86% and 95.75% respectively.According to the analysis of the probiotic characteristics,the strain significantly inhibited pathogenic bacteria such as Escherichia coli,and it has good acid resistance,bile salt resistance and strong hydrophobicity.2.Through high-throughput sequencing of L.brevis PDD-5,a total of 1158 genes were significantly expressed according to the reference gene control of L.brevis ATCC14869,among which 605 genes were significantly up-regulated and 553 genes were significantly down-regulated,and the signal pathway was enriched through KEGG database.The purine metabolic pathway was selected for analysis,and it was speculated that L.brevis PDD-5 may efficiently metabolize purine nucleoside,L.brevis PDD-5 may not contain genes that had similar functions to xanthine oxidase,and the end product of metabolism,xanthine,was metabolized into bicarbonate ion and ammonia instead of a large amount of uric acid.3.The hyperuricemia rat model was established to verify that the L.brevis PDD-5intervention group can play a protective role in the hyperuricemia animal model,and can effectively indirectly alleviate the symptoms of uric acid nep Hropathy caused by hyperuricemia.Results show that inosine and guanosine in the intestine were absorbed and degraded by the intervention of L.brevis PDD-5,leading to a significant reduction in uric acid levels in the blood of the rats,so urate was reduced in the rat kidneys.Therefore,reduced urate crystals and decreased protein expression levels of NLRP3,ASC and caspase-1 in rat kidneys lead to decreased expression of IL-1?,and ultimately inhibit NLRP3 inflammatory corpuscle to alleviate uric acid nephropathy induced by hyperuricemiaIn summary,this study found that the screened strains with strong ability to absorb inosine and guanosine can absorb and degrade the substances that are easy to form uric acid into carbonate ions to assist in reducing uric acid in purine metabolism pathway.It was proved by animal experiments that the protective treatment of hyperuricemia and uric acid nephropathy caused by the strain was feasible.All of these can provide theoretical basis for the follow-up research on microecological remissions for hyperuricemia,gout diseases and lactic acid bacteria.
