Fluorescence probe imaging provides very low detection limits and reliable immediate results.The development of fluorescence spectroscopy makes fluorescent chemical sensors not only used in vitro,but also in vivo.Fluorescence probes have advantages such as high sensitivity,high convenience,non-intrusive and high-time resolution,and fluorescence probes emitted by long wavelength emission and near-infrared light have more excellent characteristics,lower biological background fluorescence interference,less light hurt,and deep tissue penetration ability.Two-photon fluorescence probes have longer excitation wavelength,the larger imaging signal-to-noise ratio,smaller fluorescent background interference,which other traditional probes are incomparable.Reactive oxygen species(ROS)have proven to play an important role in various physiological and pathological processes.Reactive sulfur species(RSS)are regarded as the central medium of cellular regulatory function such as angiogenesis,apoptosis,signal transduction,and neuromodulation.Therefore,there is a very important research demand for the fluorescence imaging of ROS and RSS.Fluorescence probes can be useful to reveal the biochemical pathways and physiological effects of ROS and RSS.Fluorescence probes are based on the conjugate frame of the probe in this paper.Through innovative synthetic strategy,the D-?-A structure and the organelle targeting function groups design synthesized a new type of spirobifluorene-based two-photon fluorescence probe SPF.SPF-1 is a two-photon fluorescent probe for detecting H2O2for detecting a dual photon fluorescent probe.The probe SPF-2 is for cysteine(Cys)The probe SPF-3 is for rapid detection of H2S.And the fluorescence probe SPF-4 is for detecting H2S.A total of four long-wave transmitted two-photon fluorescence probes based on spirobifluorene.New tools and methods are proposed for fluorescence probe imaging ROS and RSS.Among them,the probe mother SPF has two-photon absorbing properties,longer fluorescence emission wavelength,extremely large Stoke shift(140 nm)and lysosomal targeting ability,Fluorescence probes derived from probe parent SPF also have its excellent performance.The probe SPF-1can specifically detect H2O2,which can eliminate interference of other oxidative substances,and the detection limit is 57 n M.The probe SPF-2 can specifically detect cysteines,not subject to other RSS interference.The probe SPF-3 can achieve extremely rapid detection for H2S(in 10 seconds),and the limit of detection can reach20 n M.The probe SPF-4 also has the specific detection ability of H2S and excellent detection performance.This paper expands the type of fluorescent probe based on the spirobifluorene and proposes a new tool and strategy for fluorescence probe imaging.
