Type I collagen has good biocompatibility and biodegradability,which can promote cell proliferation.It is an ideal biomedical material,which has a wide range of applications in wound dressing.The content of collagen in rabbit skin is high,and there is no limitation of traditional mammal in religion and disease.At the same time,the functional properties of rabbit skin collagen is superior to aquatic collagen.In this paper,rabbit skin collagen(RSC)was extracted from rabbit skin by acid method,and the extracted process of RSC was optimized by response surface.The results showed that the extraction rate of was 69.18% when the extraction temperature was 25 ?,the p H of the solution was 2.5,and the extraction time was 3 days.After that,the physical and chemical properties of RSC were characterized.The results showed that RSC was type I collagen which complete triple helix structure.Then the RSC was used as raw material to prepare RSC hydrogel by dialysis.The effects of mass concentration,p H and temperature of RSC solution on RSC hydrogel were investigated.And a mathematical model was established based on the change of storage modulus and loss modulus of RSC solution from liquid to gel state,then the conditions of gelation of RSC solution were explored.In order to improve the antibacterial properties of RSC hydrogel,RSC/O-CMC composite hydrogel was prepared by adding Ocarboxymethyl chitosan(O-CMC)in RSC solution.Meanwhile,the effects of O-CMC on the physicochemical properties of the composite hydrogels were studied.The results showed that the composite hydrogels had better antibacterial and rheological properties than the RSC hydrogel,and maintained the excellent properties of gel,which had a broad application as antibacterial hydrogel dressing.RSC nanofiber membrane was also prepared by electrospinning whihc RSC was the raw material,and the spinning process was optimized by single factor experiment.The results showed that when the concentration of was 9%,spinning voltage was 20 k V,spinning speed was 0.8ml/h and the receiving distance was 15 cm,the nanofibers with average diameter of 776 nm could be obtained.By adding O-CMC into RSC spinning solution,the nanofibers obtained excellent antibacterial properties.Because the carboxyl group in O-CMC formed amide bond with the amino group in RSC,the mechanical and thermal properties of antibacterial RSC nanofiber membrane were improved compared with RSC nanofiber membrane.In addition,the antibacterial RSC nanofiber membrane had good hydrophilicity and water absorption,which met its requirements as a antibacterial coating dressing.
