The dementia is a neurodegenerative disease,which is a common disease and frequently encountered disease in the elderly,including Alzheimer's dementia(AD)and Vascular dementia(VD),mixed dementia and other dementia.Alzheimer's is the most common type of dementia.AD damage brain cells,leading to memory,thinking,behavior disorders,and influence work,family and social relations.Finally affect the basic daily life ability(ADL),and lead to patients,families and society to bear a heavy economic burden.In our country,the prevalence of dementia is 2-5%.With the increase of aging population,especially in urban areas,dementia is on the rise.The number isestimated to be 4-5 million in China.Jinsiwei is a clinical experience prescription of Dongzhimen hospital,which is composed ofginseng extract,cistanche,etc.Pharmacological experiments show that it can promote the expression of hippocampal neurons ChAT and NGF,maintain normal function of cholinergic nerve.It can improve learning and memory function ofalzheimer's disease model ratsconsequently.Jinsiwei can significantly improve the learning and memory function of sporadic senile dementia model rats.It is clinically used to treat alzheimer's disease,curative effect is remarkable.So we want to make it granules.Ginseng extractof prescription can improve the body immunity,anti-aging,protection of the central nervous system.The most components are water soluble,of which the oral bioavailability is low.The essential oil in the prescription,such as acorus tatarinowii,have the function of the aromatic switched on,andpromoting the absorption of other drug ingredients.Study the intestinal absorption of before and after the main ingredient in prescription compatibility by using international unidirectional intestinal perfusion model rats in vivo.It has very important guiding significanceTo study the main composition of prescription compatibility and dosage form design.This paper studied the preparation process of Jinsiwei granules and the promoting absorption of ginseng extract after compatibility with acorus rhizoma.Details are as follows:1.The study on preparation of compound JinsiweiThe determination method for echinacoside,acteoside and 3,6'-Disinapoyl sucrose by HPLC method was firstly established.Comparing the processthat extract the whole prescription with water and the processthat extract and alcohol precipitationby combining with index component content and mice platform experimental results.By the result,alcohol precipitation can low impurity content and make effect increased.So the water extractions of prescription group refined by alcohol sink.According to the effective parts of drugs inprescription,five process routesare designed.These are compared by combining index component content and pharmacology experimental results.It shows that the proces route 4 is more advantage in pharmacological experiment results and the cost of production.That is calamus and turmeric are extracted with 95%ethanol and water extract-alcohol precipitation of other ingredientsThen the the extraction technology of water extraction and alcohol extraction group were studiedrespectively with orthogonal test method.Research on alcohol sink and drying process by using the single factor investigation method.And the forming technology of Jinsiweigranules was determined.Finally compound preparation process was determined as follows:Take medicinesaccording to the prescribed proportion,extract of two hourswith eight times the amount of water for the first time,and then filter it.Extract of two hours with six times the amount of water for the second time,filter it again.Merge these two filtrate.Enrichment to relative density 1.10(20?).Adding ethanol to make the alcohol concentration is 50%.Place at room temperature for 12 h.Filtering,take the supernatant.Decompression drying 6 hat 70?.Take rhizome and radix curcumae according to the prescribed proportion.extract of 1.5hwith six times the amount of 95%ethanol for the first time,and then filter it.Extract of 1.5h with five times the amount of 95%ethanol for the secondand the third time,filter rhizome'sfunction ofpromoting absorption to ginseng extractThe determination method for Ginsenoside-Rg1,Ginsenoside-Re andGinsenoside-Rd by HPLC method was firstly established.Use simple HPLC method to determinatethese three kinds of saponins in biological samples,andmethanol is choosedto precipitate protein in the samples,which make thepretreatment of simple rapid.Compared with the blank biological samples,the separation of these three kinds of saponins in biological samples is good,without interference from the metabolites and endogenous substances.After verification of methodology,linearity,precision,recovery rate and stability all met the requirements.This method can satisfy the requirements in studying rhizome's function of promoting absorption to ginseng extract.In this study,weused ICR male mice as model animal and choose one-way intestinal perfusion model.We studied the absorption ofdifferent concentrationsof ginseng extract in various intestinal segments,andthe absorptionafter compatibility calamus.The weight method was adopted to Calculate the absorption rate constant(Ka)and apparent absorption coefficient(Papp).Then carry on the one-way ANOVA with SPSS 17.0.The results showed that,within the scope of the mass concentration of 0.5?2.0 g·mL1.There was no significant influence in the small intestine absorption ratefor thedifferent concentration of these three ginsenosides,which reminder that the diffusion absorption mechanism was passive.It also showed that Ginsenoside-Rg1,Ginsenoside-Re andGinsenoside-Rd was absorbed the best in the duodenum.It can make Ka values increased 7.8,6.3,3.8 timesof these three ginsenosides respectivelyafter compatibility calamus extract.The effect to promote the absorption was very obvious.
