Background:Diabetes is due to glucose tolerance decreased and blood glucose level abnormally increased that cause systemic metabolic disorder.With the accelerating pace of social life and increasingly diverse lifestyles,in recent years,a sharp increase in the population of patients suffering from high blood sugar and diabetes.It brings a lot of pain to the patients,seriously affecting people's physical and mental health.Diabetes health have accounted for about 8%of the total budget in developed countries,and the United States average annual cost of diabetes for up to $ 100 billion.For decades,many countries have made unremitting research and development.Currently,dozens of Chinese and Western medicine have been used in the clinical treatment,but patients with long-term use of Western medicine will have kidney,liver,heart,spleen,stomach and other organs' side effects.Once the drug withdrawal,the relapse rate is high.Patients eventually have to rely on large doses of insulin injections or large doses of drugs.They not only have toxic side effects,but easy to produce high blood pressure and stroke,etc.They bring greater harm to the patients.Therefore,in recent years,researchers have developed a number of new compound traditional Chinese medicine which can reduce blood glucose.The compounds were deeply loved by people with diabetes.Research purposes:The purpose of this study is to provide a compound medicine that effective in treatment,easy to prepare and take,and non-toxic side effects.In accordance with the principles of Chinese medicine Medicinal and Edible and reference the modern research achievements,a Chinese compound medicine called Cang Ju tablets was prepared.Cang Ju tablets can reduce blood glucose,improve glucose tolerance indicators and prevent diabetes.Furthermore,it is a meaningful exploration for the development of new drugs.Research methods:In the preparation of extraction,in order to optimize the extraction method,orthogonal test was used.In the process study,we preferred wetting agent dosage and the ratio of material through the contrast test.In the study of quality standard,we determinate the contents of the target components by HPLC.Finally we developed the quality standards of Cang Ju tablets.We investigated Cang Ju tablets' side effects by acute toxicity test in mice.Cang Ju tablets'?-glucosidase inhibitory activity were investigated by ?-glucosidase inhibition test.Flavonoids' structure-function relationship with?-glucosidase inhibitory activity were studied.In Schwann cell viability experiments,Schwann cells were cultured and modeling.Using Giemsa staining,DAPI staining,AO/EB staining and flow cytometry,we explored Schwann cells'morphology and apoptosis mechanisms with Cang Ju tablets.And finally,we investigate the impact of glucose tolerance by Cang Ju tablets through the test in rats and mice respectively.Results:By orthogonal test,the best extraction process was determined as follows:adding 10 times amount of solvent and dipping extracted twice with each time 45 days.Combined extracting solution after extraction,we continue.concentrated them under reduced pressure.Finally they were dried to constant weight under reduced pressure,cooling and set aside.After process study,the best process optimization was determined as follows:the compounds according to the proportion of 6:1 mixed with excipients.The wetting agent according to the ratio of 1:12 mixed with raw material.By HPLC,a quality standards of Cang Ju tablets was developed.The results show that the qualitative and quantitative method is accurate,reproducible,easy and useful.It can be the quality control standards of Cang Ju tablets.Through the observation of acute toxicity in mice and pathological section,we obtained that Cang Ju tablets has lower oral dose toxicity and can be safely use.Cang Ju tablets' ?-glucosidase inhibitory activity experiments showed good ?-glucosidase inhibitory active and have some structure-activity relationship with flavonoids.The more hydroxyl group they have,the inhibitory effect be stronger.Through Schwann cell viability experiments,we confirmed that Cang Ju tablets have a significant protective function for Schwann cells.They can effectively suppress high glucose leads to cell death,and mitigate damage caused by changes in cell morphology.It showed that certain substances of Cang Ju tablets could inhibit the apoptosis of Schwann cells to exert their protective activity.Finally,the impact of glucose tolerance showed Cang Ju tablets have no significant efect on fasting blood glucose in normal rats or mice,but they can enhance glucose tolerance in normal rats and mice.Conclusions:In this study,by means of extraction,cell and enzymatic activity,animal methods,we conduct extraction and activity experiments with the prescription components.With the application of modern pharmacodynamic model,we conduct pharmacodynamics,toxicology evaluation,and compressed them into tablets for easy taking.It is meaningful for the development of new drugs and.some experience is gained.This study laid the foundation of clinical prescription medicine treat for diabetes developed into new drugs.
