Tumor metastasis is a complex process involving multiple steps.Although early diagnosis and comprehensive treatment of tumors have made significant progress, metastasis is still an important cause of death for cancer patients. Therefore,it is urgent to look for effective solutions of inhibiting tumor metastasis. Our collabotator Professor Zhou has been focusing on the functions of the novel tumor suppressor gene JWA for many years, and has achieved a series of new findings, his research group found that activating the JWA gene can inhibit tumor metastasis. They have screened many small molecule compounds that can activate the expression of JWA gene.On the basis of that,Our research group selected three pre-selected compounds(JAC-1,JAC-4 and JAC-8)to synthesize, and JAC-4 was derivatized, and Professor Zhou research group evaluated the bioactivity of JAC-4 and its seven derivatives(JAC-4-1-JAC-4-7).The results show that in Western Blot,JAC-4 activating JWA gene expression activity was better than its seven derivatives and in cell scratches experiments,JAC-4 showed a strong inhibition of cell migration activity. This indicates that the structure of 1,4-benzodioxane on the left side of the JAC-4 and the structure of methylpyridine on the right side of the JAC-4 is essential for activating JWA gene, changing 1,4-benzodioxane and methylpyridine will all affect its activation of JWA gene.On the basis of this, other members of the research group are designing and synthesing JAC-4-based biological probe,in order to find the specific target which inhibit tumor metastasis in JWA gene pathways.
