Objective:To detect the expression of heat shock protein 27(HSP27)and heat shock protein 70(HSP70)in Posner-Schlossman syndrome(PSS)patients at the onset of the disease.It is planned to explore the correlation of HSP27 and HSP70 in the pathogenesis of PSS.Methods:This study is divided into two parts.The first part is the study about the mRNA expression of HSP27 and HSP70 in peripheral blood mononuclear cells(PBMC)of PSS patients.Peripheral venous blood samples of 43 PSS patients as case group was collected from Shenzhen Eye Hospital,while the peripheral venous blood samples were obtained from 61 unrelated healthy blood donors as healthy control group.Total RNA was extracted from PBMC in venous blood.The relative expression level of HSP27 and HSP70 mRNA were detected by real-time quantitative polymerase chain reaction,in order to investigate the correlation of the expression of HSP27 and HSP70 mRNA during the onset of PSS;The second part aims to analyzes the expression of HSP27 and HSP70 in plasma of PSS patients.The plasma sample of 71 PSS patients as case group was collected from Shenzhen Eye Hospital,while the plasma sample of 99 unrelated healthy blood donors as healthy control group.The plasma contents of HSP27 and HSP70 was determined by double antibody sandwich ELISA.Analyzes the correlations between the relative expression levels of HSP70 mRNA and the HSP70 in the plasma by Spearman Correlation analysis.Result:1.The relative expression level of HSP70 mRNA in PBMC of PSS patients was significantly higher than that the healthy control group,and the foldchange was 2.52(Z=-6.097,P <0.001).);The relative xpression level of HSP27 mRNA peripheral blood PBMC of PSS patients was not statistically significant from the control group(Z=-0.977,P> 0.05).2.Plasma level of HSP70 in PSS patients was significantly lower than the control group(Z=2.930,P < 0.05);While there was no significant difference in plasma level of HSP27 between the PSS and control group(Z=-0.642,P> 0.05).3.The plasma level of HSP70 protein was positively correlated with the relative expression level of HSP70 mRNA in peripheral blood PBMC in PSS patients(r= 0.519,P<0.05).Conclusion:1.The mRNA and protein levels of HSP70 are significantly increased during the onset of PSS,which is related to the onset of PSS and may be involved in the occurrence and development of PSS.The mechanism for its involvement needs further study.2.There may be no significant difference in the expression of HSP27 mRNA in PBMC and HSP27 in plasma during the onset of PSS,suggesting that HSP27 is not related to PSS.
