| ObjectiveDepression is a kind of emotional disease with persistent depression and loss of interest as the main symptoms of the disease.It showed a high incidence rate,high recurrence rate and high suicide rate characteristics.It cause serious economic burden and mental stress to patients,their families and the society.Research on the pathogenesis of depression involves many aspects.But so far it has not been completely clear.These all explain the complexity of the pathogenesis of depression.Studies have found that long-term and frequent use of antidepressants can bring many side effects,including cardiovascular disease,liver and kidney damage,etc.However,the acupuncture treatment of depression is effective and safe.There are many studies on electroacupuncture in the treatment of depression,but its mechanism is still not fully elucidated.The purpose of this study was to investigate the effects of electroacupuncture on depressive symptoms in chronic stress model rats and the changes of P11,tPA and BDNF in the brain regions of the central raphe nucleus,observe and try to explain the relationship between them,further study the possible pathways of electroacupuncture in treating depression,and provide scientific basis for the research on the mechanism of acupuncture in treating depression by establishing chronic unpredictable mild stress(CUMS).MethodsThiry-two healthy male SD rats were randomly divided into four groups:normal group,model group,drug group and electroacupuncture group,with 8 rats in each group.After adaptive feeding,the model group,the drug group and the electroacupuncture group received chronic unpredictable stress stimulation for 28 days and were given 7 different kinds of stimulation.One kind of stimulation was randomly taken every day,but the same stimulation could not occur continuously and each kind of stimulation was at least 3 times.The normal group was raised without stimulation.The model group received only stress stimulation.In the electroacupuncture group,the operation of baihui acupoint and yintang acupoint was carried out after 1 hour of stress.The drug group received fluoxetine intervention after 1 hour of stress.No intervention was performed in the normal group.The rats in each group were tested for body mass and sugar water preference on the first day and the 28th day of the experiment,respectively,to observe the behavioral changes of the rats.On the 28th day of the experiment,the brain was taken from the severed head,and the expressions of P11,tPA and BDNF in the raphe nucleus were studied by immunofluorescence staining.Reverse transcriptase polymerase chain reaction(RPCR)was used to detect the mRNA contents of P11,tPA and BDNF.Results1.The body mass test results showed that the body weight of the model group after the treatment of drugs and eletroacupuncture was significantly higher than that of the model group(P<0.05).2.The sugar water preference experiment results showed that the sugar water consumption of the model group was significantly lower than that of the normal group(P<0.05),while the sugar water consumption of the drug group and the electroacupuncture group was higher than that of the model group(P<0.05).3.Immunofluorescence results showed that tPA,P11 and BDNF were all expressed in the raphe nucleus.The positive cells and staining intensity of the three indicators in the model group were significantly lower than those in the normal group,while the expression degree of the drug group and the electroacupuncture group was higher than that in the model group.4.PCR results showed that the mRNA expression of P11,tPA and BDNF in the model group was significantly lower than that in the normal group(P<0.05),while the expression of P11,tPA and BDNF in the model group after electroacupuncture and drug treatment was significantly higher than that in the model group(P<0.05).Conclusion1.The weight and sugar water consumption of the depressed model rats after electroacupuncture and drug treatment were higher than that of the model group.2.The expression of P11,tPA and BDNF positive cells in the brain region of the raphe nucleus of the depression model rats treated with electroacupuncture was higher than that of the model group,and the mechanism of electroacupuncture affecting the occurrence of depression and the treatment of depression may play a role by affecting the p11-tPA-BDNF pathway. |
