OBJECTIVE:This study used the expression of MYO gene in children with plateau heart disease to preliminarily explore the abnormal expression and pathogenesis of MYO gene in Tibetan children with plateau heart disease,aiming to explore the evolution of children with plateau heart disease at the genetic level.Provide basis for clinical treatment of sexual heart disease,prognosis evaluation and further gene therapy.Methods:1.The subjects of this study are 20 Tibetan children of different genders and different ages from 3-14 years old who meet the inclusion criteria for high altitude heart disease.2.Collect 3ml whole blood from children with plateau heart disease who meet the inclusion criteria,store it in an anticoagulation tube,put it in a-4? refrigerator,extract DNA,store the DNA in a special refrigerator at-50?,and then send it for inspection.3.When sending the DNA for inspection,put the DNA in a special refrigerator at-20?,and then use the method of whole exon gene sequencing(exon sequencing refers to the use of sequence capture technology to capture and enrich the DNA of the exon region of the whole genome.Genome analysis method of throughput sequencing)detected the MYO gene.Results1.This study collected a total of 20 samples of children with plateau heart disease,8infants,6 preschool children,and 6 school age children,including 12 male children and 8female children.2.In the DNA quality test,there are 18 A and 2 C.After consulting relevant experts,the quality is eliminated as C,and all 18 have passed the test standards.3.The DNA test results of 18 cases showed MYO sequence.Conclusion1.MYO gene expression exists in children with high altitude heart disease.2.According to the above studies,it is proved that the expression of MYO can be detected simultaneously during the clinical diagnosis of children with high altitude heart disease,so as to judge the children with high altitude heart disease,and on this basis,carry out research on the evolution mechanism,which is the molecular Targeted therapy plays a certain role in promoting.3.The study believes that the expression of MYO gene is of great significance in the pathogenesis of children with high altitude heart disease,and the expression and regulation of related genes in MYO genes may become a new research direction in the etiology of children with high altitude heart disease.
