Objective:To investigate the morphological and quantitative changes of oligodendrocyte precursorcells(OPC)and the transcription of oligodendrocytes atdifferenttime points afterthe intervention of high-frequency repeated transcranial magnetic stimulation(rTMS)in the ultra-early period afterfocal cerebral ischemia in rats the expression of factor(Olig1),myelin basic protein(MBP)and NG2 glial cells.Methods : Sixty-three adultmale Sprague-Dawley rats were randomly divided into model group(n=21),rTMS group(n=21),sham operation group n=21),and the three groups were divided into 1 day,3 day,and 10 day afteroperation.7 only.The model group and rTMS group were used to prepare transientmiddle cerebral artery occlusion(tMCAO)models with thread embolization.The model group and sham operation group recovered spontaneously aftersuccessful modeling,and the rTMS group was treated with high-frequency rTMS every 24 hours aftersuccessful modeling,and the intervention was for10 days.The brains of each group were taken on the first1,3,and 10 days afterthe successful modeling,and the proliferation of oligodendrocyte precursorcells and myeloid were observed by immunohistochemical staining of NG2 glial cells and myelin basic protein(MBP).The degree of sheath damage;Western blotto detectthe expression of olig1,MBP,NG2.Results : rTMS reduced the neurological deficitscore(Modified Neurological Severity Score,m NSS)aftertMCAO.Aftertreatment,the neurological deficitscore of the rTMS group was lowerthan thatof the model group(P < 0.05);rTMS promoted the proliferation and differentiation of OPC aftertMCAO.The average expression level of NG2 in the rTMS group was higherthan thatin the model group on the 1,3,and10 days(P<0.05);the average expression level of Olig1 was higherthan thatin the model group on the 3rd and 10 th days(P<0.05).rTMS promoted the repairof myelin sheath aftercerebral ischemia.The protein expression of MBP in the model group was always lowerthan thatin the sham operation group(P<0.05).Aftertreatment,the expression of MBP protein in the rTMS group was higherthan thatin the model group on the 10 th day.(P<0.05).Conclusion:Interventional high-frequency repetitive transcranial magnetic stimulation in the acute phase afterischemic stroke can reduce the degree of myelin damage and promote OPC proliferation and myelin repair.
