Purpose:Based on NF-?B signaling pathway,by observing the effects of Forskolin(c AMP activator),PDTC(NF-KB inhibitor),H89(PKA inhibitor)and ephedra asarum aconite decoction on the expression of p-CREB,AQP5 and NF-?Bp65 in rats with allergic rhinitis,further study the effects of ephedra asarum and aconite decoction on the expression of p-CREB,AQP5 and NF-?Bp65 in the treatment of AR,and explore the mechanism of ephedra asarum and aconite decoction on AR,so as to provide a theoretical basis for the pathogenesis of allergic rhinitis in Chinese medicine such as Yang Qi deficiency and water and drink withdrawal,and further reveal the rationality of treating AR by drinking in Chinese medicine in orderto provide a reasonable direction for the clinical treatment of AR.Material and method:48 healthy SPF male wistar rats were divided into 6 groups by randomized bocks design: normal group,AR model group,Forskolin group,H89 group,PDTC group and the Mahuang Xixin Fuzi Decoction group,8 rats in each group.The AR rat model was made by the method of whole body sensitization and local attack.AR rat model was made by whole-body sensitization and local attack of egg protein.Normal control group and model control group were given normal saline gavage per 1ml/100 g.Forskolin intervention group was intraperitoneally injected with 5mg/kg/d Forskolin;H89 intervention group was intraperitoneally injected with H89 at 5mg/kg/d;PDTC intervention group was intraperitoneally injected with 50mg/kg/d PDTC;The ephedra asarum and aconite decoction group was given 2.1ml/1kg of the tested drug decoction(equivalent to 2.1g crude drug)by intragastric administration.After the last stimulation,the expressions of AQP5,NF-?Bp65and p-CREB in the nasal mucosa of the anesthetized rats were observed after death.The results were analyzed statistically.Results : The expression of p-CREB and AQP5 in the model group decreases and it increases that the expression of p-CREB and AQP5 in the Forskolin group,PDTC group,the Mahuang Xixin Fuzi Decoction group,while the PKA inhibitor H89 can decrease the expression of p-CREB and AQP5.The expression of NF-kappa B p65 in the model group increases and the expression of NF-kappa B p65 in the Forskolin group,PDTC group,the Mahuang Xixin Fuzi Decoction group decreases,while H89 can increase the expression of NF-kappa B p65.Conclusion : The mechanism of treating allergic rhinitis by the Mahuang Xixin Fuzi Decoction group may increase the expression of AQP5 by inhibition of NF-kappa B signaling pathway and exciting c AMP-PKA signaling pathway.
