The genital tract of female animals is more susceptible to pathogenic microorganisms because of it open directly to the body surface.In animal production,the infections of genital tract caused by Gram-negative bacteria and the reproductive disorders caused by Gram-negative bacteria are the main reasons that lead to the decrease of female animal reproductivity.Lipopolysaccharide(LPS)is a component of the cell wall of Gram-negative bacteria,as a pathogen-associated molecular pattern of Gram-negative bacteria,is usually used to simulate inflammatory response instead of Gram-negative bacteria.Cyclosporine A(CsA),a common immunosuppressive agent,is often used in rejection after organ transplantation.However,the knowledge on the effect of CsA on the expression of chemokine,and the effect of CsA on chemokine and immune cell recruitment in genital tract of female rabbits induced by LPS still was unkown.Our objective is to explore the effect of CsA on the expression of chemokine and the recuriment of immune cell.The concentration of chemokine in the serum and uterus,and the expression level of chemokine in different segments of genital tract was examined,and then the recuiment of CD4~+/CD8~+T cells and CD11b~+neutrophils in the uterus was examined after the female rabbits were treated with or without CsA under the status of LPS-induced inflammatory response.Results,(1)LPS induced the expression of IL-6 in the uterus,and CsA inhibited the LPS-induced up-regulation of IL-6 expression in the uterus and CsA induced the expression of anti-inflammatory cytokine IL-10 in the uterus.(2)LPS significantly down-regulated the expression of CCL4 in the cervix,and CCL14 and CCL3 in the uterine body and isthmus,and MCP-1 in the isthmus.CsA-treatment significantly upregulated the expression of CCL4 in the uterus and significantly down-regulated the expression of MCP-1 in the isthmus.In LPS+CsA group,the expression of CCL24 in the cervix and uterus body was significantly lower than that in LPS-treatment group,while the expression of CCL4 in the isthmus was significantly higher than that in LPS-treatment group.(3)LPS could significantly up-regulate the recruitment of CD4~+T cells in the serosa of the uterus and the recruitment of CD11b~+neutrophils in the endometrial layer.The recruitment of CD8~+T cells in LPS+CsA group was significantly lower than that in LPS group.The results suggested that LPS induced uterine inflammation of female rabbit,while CsA could inhibit the inflammatory response induced by LPS.LPS and CsA changed the expression of chemokines,and the changes depends on segment of the reproductive tract.LPS induced the recruitment of CD4~+/CD8~+T cells and CD11b~+neutrophils in the uterus,while CsA only inhibited the recruitment of CD8~+T cells in the uterus.
