| Background:Aedes albopictus,as an invasive species,can transmit a variety of viral diseases.This species successfully survived developmental and environmental stress by its unique physiological features coordinating metabolic pathway.Understanding the metabolic changes in each developmental stage and deciphering regulatory mechanism coordinating energy metabolism can contribute to the implementation of metabolism-based methods controlling mosquito.Objective:Revealing metabolic differences by finding differential metabolites and metabolic pathways in different developmental stages of Ae.albopictus using 1H-NMR based metabolomics.Exploring how hormones regulates the sugar metabolism of Ae.albopictus.Methods:(1)Metabolites of Ae.albopictus were detected by 1H-NMR based metabolomics,the collected free Induction Decay(FID)signal were processed by Chenomx NMR suit to identified and quantified metabolites in each sample.(2)Statistical analysis was performed to screen differential metabolites and differential metabolic pathways using MetaboAnalyst4.0 analysis platform.(3)Transcriptome data of Ae.albopictus were annotated[1]expressional dynamics of hormone genes and carbohydrate metabolism genes throughout Ae.albopictus’developmental cycle were analyzed,and further verified by qRT-PCR;correlation analysis between carbohydrate metabolism genes and hormones were performed.Correlation analysis between metabolites and hormones were carried out based on metabolomics’ data.(4)Different concentretion of juvenile hormone and 20-hydroxyecdysone were used to treat C6/36 cells,fat bodies and 1-2 instar larvae of Ae.albopictus for 24 hours.The appropriate hormone final concentration was used to deal with C6/36 cells for different durations.The samples above were collected to perform qRT-PCR and detect cellular glycogen.Results:(1)Different mosquito developmental stages displaced respective metabolism feature,and some metabolites and metabolic pathway showed stage-specific characteristic.(2)Expressional dynamics of carbohydrate metabolism genes,hormone genes,hormone receptors genes throughout the Ae.albopictus’ developmental cycle were showed.(3)Juvenile hormone has significant positive correlation with glucose,maltose and 19 carbohydrate metabolism genes;ecdysone has significant positive correlation with amino acids,and a negative correlation with trehalose;hormone receptors had no correlation with any genes nor metabolites discussed above.(4)Hormone stimulation experiments showed that juvenile hormones had different effects on the glucose metabolism of C6/36 cells,fat bodies,and instar larvae,but significant changes were not found in the ecdysone-stimulated group.Conclusion:(1)Metabolic profiles in developmental life cycle of Ae.albopictus displayed a highly stege-specific characteristic;(2)Juvenile hormones were positively correlated with glucose,maltose and 19 carbohydrate metabolism genes,ecdysone were positively correlated with amino acids,and negatively correlated with trehalose;(3)Juvenile hormone played a vital role in carbohydrate metabolism of Aedes albopictus. |
