In recent years,with the vigorous development of petroleum exploration and petrochemical industry,in the process of exploration,development,storage and transportation,processing,storage and sales of petroleum and products,a large number of petroleum and products have been caused due to backward technology and non-standard operation.The leakage has seriously endangered the ecological environment of the soil.The remediation of petroleum hydrocarbon-contaminated soil has become a research hotspot at home and abroad.The commonly used remediation methods include physical methods,chemical methods,biological methods,etc.Among them,microbial remediation methods have high remediation efficiency,low cost,no secondary pollution,The technical advantages such as small impact on the soil ecological environment are considered to be a sustainable means of dealing with contaminated soil from large-scale oil fields.The content of petroleum hydrocarbons in petroleum hydrocarbon-contaminated soils is the main indicator of the degree of soil pollution and an important basis for determining the effect of soil remediation.This thesis takes the oil-contaminated soil from Gudao Oil Production Plant in Dongying Shengli Oilfield as the main research object.Through the comparison of various analysis methods,a method for accurately determining the concentration of petroleum hydrocarbons in the process of microbial remediation of contaminated soil is determined.Using the growth conditions of microorganisms,determine The influencing factors of microbial inoculum remediation of oil-contaminated soil;the method of orthogonal test is used to determine the optimal process conditions for microbial remediation of oil-contaminated soil;the best process conditions obtained are used to construct the degradation kinetics of microbial remediation of petroleum hydrocarbon-contaminated soil Model.The result is as follows:(1)The content of petroleum hydrocarbons in the process of microbial remediation of contaminated soil was determined by gravimetric method,UV spectrophotometry and gas chromatography,and the four components of petroleum hydrocarbons were analyzed.It can efficiently and accurately measure the change of petroleum hydrocarbon content in microbial remediation polluted soil.(2)Through experiments on the process of microbial remediation of oil-contaminated soil,it was determined that temperature,soil water content,ventilation,the type and amount of soil amendments,and the amount of bacteria added are the main factors affecting the microbial remediation effect.Temperature can promote the process of microbial remediation of petroleum hydrocarbon pollution.It is advisable to keep the soil moisture content at about 20%.Regularly adding a certain amount of high-efficiency petroleum hydrocarbon degrading bacteria to the polluted soil has a significant promoting effect on the degradation of petroleum hydrocarbon pollutants;adding Corn stalks can improve soil aeration and increase the degradation rate of petroleum hydrocarbon pollutants in soil.(3)The best remediation process was determined by orthogonal experiments: adding 5% of petroleum hydrocarbon efficient degrading bacteria and 5% of corn stover,keeping the soil temperature at 25 ℃,and maintaining the soil moisture content at 15% in the early stage of remediation,The soil moisture content was maintained at 20% in the later stage of remediation.After 70 days of repair under this process condition,the degradation rate of petroleum hydrocarbons can reach 62.8%.(4)It was determined that the variation trend of petroleum hydrocarbon degradation and time in the contaminated soil was in line with the second-order kinetic model.The kinetic equation was: 1/ct=0.00354t+1/9.7,and the correlation coefficient was0.942.The petroleum hydrocarbon degrading bacteria The half-life of the agent in the repair process is 29 d.
