| Torreya grandis is an evergreen tree of Torreya in Taxaceae,which has high value for both medicine and food.T.grandis(T.grandis seed,T.grandis seed oil)was used as the research material,feed the mice with T.grandis diet,detected the serum biochemical indexes.The pathological sections of mice were observed to analyze the proportion of liver lipid droplets and the changes of intestinal villi and crypts.Combined with various omics techniques,the regulatory effect of T.grandis diet on hyperlipidemia mice was analyzed.The main results are as follows:1.T.grandis diet can regulate the blood lipid level and serum antioxidant capacity of mice.Serum biochemical indexes showed that dietary supplement of T.grandis seed could significantly improved the levels of serum TC,TG and high density lipoprotein cholesterol(HDL-C)in hyperlipidemia and normal mice(P<0.05).The levels of serum superoxide dismutase(SOD),total antioxidant capacity(T-AOC)and glutathione peroxidase(GSH-Px)were significantly increased(P<0.05).The levels of serum immunoglobulin(Ig G,Ig A and Ig M)were significantly increased(P<0.05).Dietary supplement of Torreya grandis seed oil could significantly improved the levels of TC,TG,HDL-C and low density lipoprotein cholesterol(LDL-C)in normal mice(P<0.05),significantly reduced the level of blood glucose(Glu)in normal mice(P<0.05),and significantly increasde the level of antioxidant(SOD,GSH-Px)(P<0.05).2.T.grandis diet can improved the liver and intestinal health of mice.The results of histopathological sections showed that T.grandis diet significantly reduced the proportion and area of liver lipid droplets in mice and alleviated nonalcoholic fatty liver.High fat diet significantly increased the depth of crypt in mice(P<0.05),and T.grandis seed diet significantly decreased the depth of crypt in mice(P<0.05).T.grandis diet relieved hyperlipidemia in mice through a variety of mechanisms.3.Serum metabolome analysis showed that T.grandis seeds could regulate the metabolic pathways such as blood glycerol,phospholipid,fatty acid biosynthesis and extension.The transcriptome and metabolome of liver were analyzed,which showed that high-fat diet caused abnormal lipid metabolism in mice.Torreya grandis seed oil may regulate metabolic pathways such as cholesterol,fat metabolism and absorption,amino acids and alleviate abnormal lipid metabolism in mice.4.16 S r RNA sequencing analysis showed that Torreya grandis seed diet increased the relative abundance of Lachnospiraceae,lachnospiraceae-NK4A136-group and Alloprevotella in mice.Torreya grandis seed oil diet significantly increased observed species(1508.75),Chao1(1723.39)and ACE index(1852.58)(P<0.05). |
