Solanaceae has high edible and medicinal value,and is also an important experimental material.In the reproductive growth process of Solanaceae plants,flower development is comprehensively affected by many genes,among which AGAMOUS(AG)gene plays an important regulatory role in flower and fruit development.In this study,four AG genes were obtained by transcriptome data and bioinformatics analysis,and Sn AGL5 was selected as the target gene.The function of Sn AGL5 was studied by genetic transformation,q RT-PCR and tissue section analysis.1.Bioinformatics analysis Sn AGL5 encodes 232 amino acids with MADS-box,K-box domains and two AG-motifs at the C-terminal,and has the closest relationship with MADS-BOX PROTEIN 3(Sl MBP3)in Solanum lycopersicum.2.The results of tissue-specific expression showed that Sn AGL5 was only expressed in the carpels of flowers,and the expression level of Sn AGL5 in flowers at different stages gradually increased,while it first increased and then decreased in fruits.3.The results of overexpression of Sn AGL5 in S.nigrum showed that the sepal cells of the transgenic Sn AGL5 line changed from nearly circular to long oval cells similar to the pericarp,and the sepals showed that the fleshiness gradually changed into purple-red.Through anthocyanin analysis,the anthocyanin content in the sepals of the transgenic lines was much higher than that of the wild type.
