The trichome and thorns in plants share very similar function.They all can protect plants against biotic and abiotic stresses.R2R3-MYB is the largest MYB family in plant MYB gene family,it plays many important roles in plants,including regulating the development of trichome.Caragana korshinskii Kom.has high ecological and economic value.However,it has stipular spines,which brings a lot of inconvenience in feeding,harvesting,seed collection and processing.Its economic benefits would be increased if the new germplasm of C.korshinskii without or with less or soft stipular spines could be developed.Therefore,it is necessary to verify the genes related to stipular spines formation in C.korshinskii.In this study,based on the literature and the genome data of C.korshinskii,a R2R3-MYB gene named as CkMYB61 was cloned and its function was analyzed in Arabidopsis thaliana.In addition,the transcriptome of C.korshinskii with and without stipular spines was analyzed and a set of candidate differentially expressed genes(DEGs)were selected.The main results of this study are as follows:1.CkMYB61 has an open reading frame of 1269 bp and encodes 422 amino acids.Bioinformatics analysis of the protein encoded by this gene showed that CkMYB61 was an unstable hydrophilic protein with a molecular mass of 47.12 k D,and an isoelectric point of7.12.2.The expression pattern analysis of CkMYB61 showed that the expression level of this gene was the highest in stem and the lowest in pod of C.korshinskii.3.After overexpression of CkMYB61 in Arabidopsis,it was found that the transgenic Arabidopsis plants showed a significant increase in leaf trichomes and a shorter root length.4.After analyzing the transcriptome of the section with and without stipular spines of C.korshinskii,1,252 DEGs were obtained.After further analysis,39 genes promoting and105 genes inhibiting the formation of stipular spines were selected as the candidate genes for subsequent confirmation.
