At present,the pathway of seed oil biosynthesis is clear,but the research of fatty acid synthesis regulatory network needs to be further explored.In the work,we constructed a yeast library with seed cDNA of Arabidopsis thaliana and screened the interaction protein using LEC1,an important transcription factor regulating fatty acid synthesis,as bait protein.In addition,we intend to study the interaction protein in regulating fatty acid synthesis and oil accumulation in seeds via molecular biology,genetics,biochemistry and bioinformatics methods in this project,so as to lay a foundation for further revealing the regulation mechanism of fatty acid synthesis and oil accumulation,and then provide theoretical support for molecular breeding to obtain high oil yield crop varieties.The main results of this study are as follows:1.Constructed Arabidopsis seed cDNA yeast library and screened JAZ9 protein interacting with LEC1 The seed cDNA yeast library was constructed with Arabidopsis thaliana Colombian ecotype pods as materials.The interaction protein JAZ9 was screened by using LEC1,an important transcription factor regulating fatty acid synthesis,as the bait protein.2.LEC1 interacts with JAZ9,JAZ1.JAZ9 does not interact with WRI1,FUS3 and LEC2,but specifically with LEC1.LEC1 can also interact with JAZ1.It is verified that JAZ9 can also interact with JAZ1.3.Obtained ATLEC1 and ATJAZ9 overexpression plants.The overexpression vectors of LEC1 and JAZ9 were constructed by using pcambi1300 and pcambi1301,and the overexpression plants of LEC1 and JAZ9 were obtained by transgenic technology.
