Studies On Insecticidal Constituents From Clematis Aethusifolia

Abstract/Summary:
Clematis aethusifolia Turcz.is perennial herbaceous vine,belong to the family of Ranunculacea,widely distributes in Southwest of China.This herb can used as medicines for dispelling wind and dampness,relaxing muscles and accelerateing blood circulation,and relieving pain.Different types of compounds have been separated from this genus,including saponins,flavonoids,coumarins,lignans,alkaloids and so on.This study focused on the isolation and identification of insecticidal constituents in Clematis aethusifolia Turcz.as well as the evaluation of insecticidal activity of the pure compounds.The main results are shown as follows:1.The ethanol extract of the roots of Clematis aethusifolia was isolated from co lumn choromatography on normal phase silica gel,Sephadex LH-20,reversed-phase si lica gel and semi-prepared HPLC to yield 10 compounds.The structure of isolated c ompounds were identified according to their physical and chemical properties and spe ctral data.They are belong to three lignins:(+)-syringaresinol-4’-O-β-D-glucopyranosid e(TQ-1),(+)-pinoresinol-β-D-glucoside(TQ-3)and samsesquinoside(TQ-4);three flav onoids:quercitrin(TQ-2),quercetin-3-rutinoside(TQ-7),quercetin-7-O-glucoside(TQ-8);and four saponins:hederagenin-3-O-β-D-ribopyranosyl-(1→3)-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranoside(Clematoside S,TQ-5),3-O-β-D-ribopyranosyl-(1→3)-α-L-r hamnopyranosyl-(1→2)-[β-D-glucopyranosyl-(1→4)-β-D-xylopyranosyl hederagenin](Cle matilasiandroside A,TQ-6),3-O-β-D-ribopyranosyl-(1→3)-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranosyl oleanolic acid 28-O-β-D-glucopyranosylester(Clematiganoside A,TQ-9),3-O-β-D-ribopyranosyl-(1→3)-α-L-rhamnopyranosyl-(1→2)-β-D-xylopyranosyl o leanolic acid 28-O-α-L-rhamnopyranosyl-(1→4)-β-D-glucopyranosyl-(1→6)-β-D-glucopyr anosyl ester(Huzhangoside C,TQ-10).2.The antifeedant and toxic activity of TQ-5,TQ-6,TQ-9 and TQ-10 were evaluated against third-larval of Plutella xylostella(L)and Mythimna separate(walker)using leaf discs method and diet-incorporation method in the concentration of 1 mg/m L,the results displayed the activities of TQ-5 and TQ-6 was better,with 48 h antifeedant rates of 65.32%and 63.08%,72 h mortality rates of 33.33%and 33.33%respectively.3.The antifeedant and toxic activity of TQ-5 and TQ-6 against third-larval of P.xylostella was determined by leaf discs method.The results displayed TQ-5 showed antifeedant and toxic activity against P.xylostella(L.)with AFC50 of 737.67μg/m L(24 h)and 748.28μg/m L(48 h),and LC50 of 1789.03μg/m L(48 h)and 1462.78μg/m L(72 h),respectively.As for TQ-6,the AFC50 were 884.77μg/m L and 744.24μg/m L at 24 h and 48 h,respectively,and the LC50 were 2152.51μg/m L and 1652.15μg/m L at 48 h and 72 h,respectively.4.The growth inhitibitory effects of TQ-5 gainst 3rd instar larvae of P.xylostella was performed at concentration 500,1000 and 1500μg/m L,the results showed that TQ-5induced a significant effect on the whole life cycle of P.xylostella.Compared to control,the larvae and pupae weights was drastically decreased with all treatments,the pupation and emergence of all treantment was drastically decreased,but the time to pupation and adult stage were delayed.Moreover,the inhibition was concentration-independent.The above results clarified the insecticidal active substance for saponins from C.aethusifolia,and laid a foundation for further study on the insecticidal mechanism of saponins from Clematis gainst lepidopteran pests.
