Influenced by UV rays,haze,dust and other environmental factors,cells undergo oxidative stress leads to exogenous skin aging.It manifests as skin sagging,increased pigmentation and inflammatory reactions,affects visual aesthetics and skin health.Daily chemical active substances with anti-oxidant,anti-aging,anti-inflammatory and other properties have attracted widespread attention.However,due to the disadvantages of large molecular weight,poor oil-water partition coefficient and poor stability,some functional substances are difficult to act through the skin and have low bioavailability,which limits their development and application in cosmetics.Liposomes are biocompatible and slow-release,and they can encapsulate both water-and fat-soluble compounds,allowing active substances to penetrate the skin and remain longer in skin cells.Liposomes,with unique structural advantages,are emerging carriers in the field of cosmetics.The preparation of liposomes with high-yield,fast,simple and low cost is the bottleneck in their industrial production.In this paper,three kinds of daily chemical active substance liposomes were prepared by Higee.The mechanized mass transfer was relatively uniform,and the operation was simple and easy to control.The main contents of this paper are as follows:(1)The liposomes prepared by Higee with convenient operation and short produced period of liposomes.Higee is projected to achieve industrial production of daily chemical active substance liposomes.When the high gravity level was 200,the ethanol/water feed ratio was 1/4,the cholesterol/lecithin ratio was 1/6,the Tween 80/lecithin ratio was 1/4,retinyl palmitate(RP)/lecithin was 1/15,the average particle size of RP liposomes(RP-LPs)was 207 nm,the encapsulation efficiency(EE)was about 95%.There was no flocculation or Ostwald ripening phenomenon after 200 days at room temperature.When heated at 60 °C for 500 minutes,RP was oxidized by51.57% while RP encapsulated in liposomes was only oxidized by 19.78%.The RP-LPs could effectively reduce the thermal sensitivity of RP.(2)Proanthocyanidins liposomes(PC-LPs)and vitamin C liposomes(VC-LPs)were prepared to explore the effect of the rotating packed bed(RPB)on the EE of active substances with different polarities and molecular weights.When the PC/lecithin ratio was 1/34,the EE was 91.41%,the particle size was191.5 nm.When the VC/lecithin ratio was 1/100,the EE of VC-LPs was91.72%,the particle size of VC-LPs was about 200.1 nm.The liposomes dispersed in solution without ions had the smallest particle size,which was about 129.5 nm,and the EE was over 90%.Experiments revealed RPB could prepare liposomes with high EE regardless of the polarity and hydrophobicity of active components.(3)Compared with phosphate-buffered saline,the dehydration rate of pigskin and artificial skin smeared with liposomes was significantly lower,it had barrier and moisturizing functions.PC,VC and RP exhibited DPPH and hydroxyl radical scavenging abilities.After being encapsulated by liposomes,PC and RP’s DPPH radical scavenging ability was enhanced.Liposomes are hydrophilic and compatible,they can be evenly mixed with other cosmetics by stirring or shaking. |
