| Koumiss has nourishing and hygienical function,but the development of koumiss industry is limited because of poor storage and insufficient application.In this experiment,koumiss powder was made by vacuum freeze drying method,and its physicochemical indices and functional characteristics were analyzed.By constructing CUMS(chronic unpredictable mild stress)mouse model,the effect of koumiss(powder)intervention on the behavior and intestinal flora of CUMS mice was investigated,in order to preliminarily explore the potential mechanism of koumiss(powder)in alleviating anxiety and depression,providing theoretical basis for the development and application of koumiss powder products.The main contents are as follows:(1)Optimization of preparation technology of vacuum freeze-drying koumiss powder.With the sensory score,flour yield,liquidity as evaluation index,the L9(33)orthogonal test was used to investigate the influence of concentration ratio,material thickness and freeze drying time on the quality of koumiss powder.The optimum process parameters were determined as follows:concentration ratio of 15%,material thickness of0.75cm and freeze-drying time of 24h.(2)Analysis of physicochemical indexes and functional characteristics of koumiss powder.The nutritional components(protein,fat,moisture,ash),acidity,total number of colonies,antioxidant properties and GABA content of koumiss powder were detected,and it was found that the nutritional composition of koumiss powder did not change significantly after drying,and the number of viable bacteria and antioxidant capacity were lost to a certain extent,but the content of GABA was enriched to 36.57±0.07mg/m L.It is speculated that it has the potential to relieve anxiety and depression.(3)Effects of koumiss(powder)on the behavior of CUMS mice.The differences of inflammatory factor TNF-αand behavior of mice in different treatment groups were investigated.The results showed that the intervention of koumiss(powder)could prevent inflammation properly.The behavior of mice in the model group tended to be abnormal,while the sucrose preference index of the mice in the koumiss(powder)intervention group increased,the immobility time in water was shortened,the emotional score was reduced,and the behavior gradually turned to normal,indicating that koumiss(powder)can relieve the anxiety and depression behavior of CUMS mice.(4)Effects of koumiss(powder)on the intestinal microflora of CUMS mice.The microflora composition of model group was significantly different from that of blank control group and intervention group.Mice in the model group showed a certain degree of dysbacteriosis,and the relative abundance of Alistipes and Prevotellaceae UCG001was significantly reduced(P<0.05),and the relative abundance of Lachnospiraceae NK4A136 and Lachnospiraceae UCG-001 was significantly increased.The medicine and koumiss powder intervention can significantly upregulate Alistipes and Prevotellaceae UCG001,significantly downregulate the Lachnospiraceae NK4A136 and Lachnospiraceae UCG-001,so that the flora gradually move closer to the normal mice flora structure,indicating that fluoxetine and koumiss(powder)can improve the intestinal flora structure of CUMS mice,and then play a role in alleviating anxiety and depression.In conclusion,vacuum freeze-dried koumiss powder has good quality and functional characteristics,and can properly improve the intestinal flora disorder of CUMS mice,prevent inflammation,and then improve the anxiety and depression behavior of CUMS mice. |
