| "SangHuang" is an important macro medicinal and edible fungus,and its main active ingredient polysaccharide has been proved to have various physiological activities,such as antioxidant,anti-tumor,anti-inflammation,etc.However,the utilization rate of "SangHuang"is still low.At present,most "SangHuang" products are mainly primary processing products.Therefore,it is urgent to develop "SangHuang" deep processing products.In this study,the contents of nutrients,active components and antioxidant activities of polysaccharides in the fruiting bodies of different varieties "SangHuang" were compared and analyzed,and the target strains were selected and identified by molecular biology.The extraction process of polysaccharide from Sanghuangporus vaninii(PSH)was optimized,and the monosaccharide composition and antioxidant activity were analyzed.The optimal process ratio and granulation process of Sanghuangporus vaninii polysaccharide composite chewable tablets were obtained.To provide theoretical basis and technical support for the development of Sanghuangporus vaninii polysaccharide functional food.(1)The contents of nutrients and active components and antioxidant activities of polysaccharides of the fruited bodies of five kinds of "SangHuang" were compared and analyzed.S-4 was selected as the target strain for subsequent experiments after comprehensive consideration.According to ITS sequence determination,S-4 was Sanghuangporus vaninii.(2)The extraction process of PSH was optimized by Plackett-Burman test and RSM test with the polysaccharide yield as the index.The best extraction process for the hot water extraction of PSH was 16 times of water addition,1.7 h of extraction time and-3℃ of alcoholic precipitation,and the actual yield under this condition was 4.98%.(3)The results of UV scan and Fourier transform infrared spectroscopy showed that PSH has typical absorption peaks of polysaccharides containing β-glycosidic bonds and furan rings.The heavy average molecular weight(Mw)of PSH was 2.29 ×105 Da,the number average molecular weight(Mn)was 2.84 ×104 Da,the Z-average molecular weight(Mz)was 4.67× 106 Da and the peak molecular weight(Mp)was 3.92 × 104 Da.The monosaccharide composition of PSH was determined to be mainly composed of glucose,galactose,fucose,glucuronide,ribose and mannose.(4)The scavenging ability of PSH on DPPH radical,ABTS+ radical and O2-radical was good,and the half inhibitory concentration(IC50)were 35.07,12.87 and 91.34 μg/mL,respectively.(5)The sensory score was used as the main evaluation index,and the tablet hardness,fragility and tablet weight difference were the auxiliary indexes.The formulation was optimized by single-factor and orthogonal tests.The optimal process ratios of Sanghuangporus vaninii polysaccharide complex chewable tablets were 34%PSH addition,19%microcrystalline cellulose addition,28%date powder addition,18%xylitol addition,and 1%magnesium stearate addition.The sensory score of Sanghuangporus vaninii polysaccharide composite chewable tablets was 80.25,hardness was 397.74 N,crispness was 0.28%,and the difference of tablet weight was 1.46%.(6)The granulation process of Sanghuangporus vaninii polysaccharide compound chewable tablets was optimized by single-factor test and RSM test with the index of granule yield.The optimal granulation process was 16 min drying time,63℃ drying temperature and 0.3%CMC-Na concentration,and the granule yield was 92.06%.Sanghuangporus vaninii polysaccharide complex chewable tablets retain the unique flavor of polysaccharide from Sanghuangporus vaninii with date and caramel aroma,sweet and sour,delicate taste and moderate hardness.The heavy metal content and microbial test results of the chewable tablets are in compliance with the national food safety standards. |
