Pleurotus ostreatus is a kind of edible mushroom that is welcomed by consumers.Yet the traditional method of wood cultivation has aggravated the contradiction between the mushroom and the forest.In this study,researches on the method of Pleurotus ostreatus cultivation with fresh Jujuncao have been conducted from the perspectives of cost saving,shortening of production cycle and slowing down of contradictions.The main content is as the following four parts.Firstly,three different cultivars of fresh Jujuncao,dried Jujuncao and sawdust were compared to carry out the bag-planting experiment of Pleurotus ostreatus.The extracellular enzymatic activities of Pleurotus ostreatus at different stages during bagging experiments were determined.At the same time,the contents of lignin,cellulose and hemicellulose of cultivated materials in different periods were determined and compared with the extracellular enzyme activities.Secondly,the physical index,biological efficiency,yield and nutrient content of Pleurotus ostreatus fruiting bodies in the bag-planting experiment were measured,and the fruiting bodies differences of Pleurotus ostreatus cultivated in different cultivation materials were analyzed using the grey correlation method.Thirdly,the antioxidant activity of the polysaccharide of Pleurotus ostreatus fruiting body in the bagging experiment was measured to analyze the difference of the antioxidant activity in vitro of Pleurotus ostreatus fruiting bodies of different cultivation materials.The main findings are as the followings:(1)Effects of different cultivation materials on extracellular enzyme activities of Pleurotus ostreatus at different stages In the process of Pleurotus ostreatus growing with different cultivation materials,determine activities extent of extracellular enzymes such as laccase,carboxymethyl cellulose,polyphenol oxidase and other extracellular enzymes during the three different mycelia growing periods of full bag growing stage,young mushroom stage and fruiting body maturation period.The results showed that after the mycelia were full of bags,the ligninase and cellulase activities of the Jujuncao as a cultivation material formulation were reduced,but the laccase activity was increased.With sawdust as the cultivation material formulation,the ligninase activity increased from the mycelia full of bag to the young mushroom formation stage.And the extracellular enzyme activity of different cultivation materials decreased from the young mushroom to fruiting body maturity.(2)Changes of lignin,cellulose and hemicellulose content in different stages of different cultivated materialsBy comparing the changes of lignin,cellulose and hemicellulose content of cultivated materials in different periods,and comparing the lignin and cellulase activities of Pleurotus ostreatus in different periods,the effects of different cultivation materials on Pleurotus ostreatus were further explored.The results showed that for sawdust,the content of lignin,cellulose and hemicellulose increased from the time after inoculation to the mycelia full of bag,after which the content began to decline,and the lignin content decreased rapidly;for the fresh Jujuncao after inoculation,the contents of lignin,cellulose and hemicellulose had been decreasing,and the content of cellulose decreased most rapidly,while the content of hemicellulose and lignin decreased slower than the former.The content of cellulose and hemicellulose of the hay Jujuncao had been reduced.The downward trend was basically the same as that of the fresh Jujuncao,but the lignin content increased after the mycelia was full of bag.For proteins,the changes in the protein content of the formulations of different cultivation materials were basically the same,both falling first and then increasing.For the protein content of the smut,the content of the formula with wood chips as the cultivation material was the highest,the fresh Jujuncao was the second,and the hay Jujuncao was the lowest.(3)Effects of different cultivation materials on the nutritional quality of Pleurotus ostreatus fruiting bodiesThe experiment was based on the use of Pleurotus ostreatus cultivated with sawdust and dried Jujuncao as a control group.The cultivation formula of the fresh Jujuncao was set to explore the changes in nutrient composition of Pleurotus ostreatus with different cultivation materials.The results showed that the formula with the highest content of crude polysaccharides and crude fibres is based on the formulation of fresh Jujuncao as the cultivation material;the formula with the highest content of crude protein is the formulation of wood chips as the cultivation material;the formula with the highest content of ash is wood chips as the cultivation material.(4)Effects of Different Cultivation Materials on Antioxidant Activity of Pleurotus ostreatus Fruiting BodiesThe effect of different cultivation materials on the antioxidant activity of alcohol extracts from Pleurotus ostreatus fruiting bodies was discussed.The results showed that:from the results of in vitro antioxidant,the best ability of the alcohol extracts of Pleurotus ostreatus fruiting bodies to scavenge superoxide anion radicals is wood chips as cultivation formula of Pleurotus ostreatus fruiting body;the best ability to remove DPPH is sawdust as cultivation formula of Pleurotus ostreatus fruiting body,and the best clear ability for hydroxyl radicals is the Pleurotus ostreatus fruiting body of the fresh Jujuncao as cultivation formula.
