With the continuous abuse of antibiotic drugs,the outbreak of super bacteria,the disease caused by multi-drug resistant E.coli has become a serious problem threatening the safety of human health and animal husbandry worldwide.In this study,ESBLs-producing Escherichia coli was isolated from pigs,and the resistance of Escherichia coli was analyzed from the antibacterial effect in vitro.The model of bacteremia in mice was established,and the compound rescued Ilex rotunda Thunb(Ilex rotunda Thunb + Cyperus rotundus L.+ Atalantia buxifolia(Poir.)Oliv.ex Benth.)and Astragalus propinquus Schischkin + Ilex rotunda Thunb should be divided into 2 prescriptions,9 single drug or compound medication groups were selected for routine disease observation,mortality,blood routine(white blood cells,neutrophils,monocytes,lymphocytes,platelet count,platelet pressure)Biochemical detection of mouse serum(alanine aminotransferase,glutamyl transaminase,aspartate aminotransferase,total protein,globulin,albumin,etc.),inflammatory factors(IL-1α,IL-1β,IL-6,IL-10),TNF-α,)and procalcitonin(PCT)and pathological anatomical maps were used to analyze the mechanism of action of different drug groups in the treatment of E.coli-induced mouse bacteremia.Finally,the group with the lowest mortality rate and the Ilex rotunda Thunb + Cyperus rotundus L.and the mouse group of the blank group and the model group were selected for differential proteomics analysis.The main experimental results are as follows:1.Clinically collect pathological data and isolate E.coli producing extended-spectrum β-lactamase(ESBLs).The study found that the ESBLs-producing Escherichia coli is multi-drug resistant Escherichia coli,and its sensitivity to ceftriaxone sodium,amoxicillin,sulfamonomethoxine,florfenicol and other antibiotics is much higher than international standards,but rescue The boiled liquid has a certain in vitro antibacterial effect on the ESBLs-producing Escherichia coli,and the MIC value of the rescued boiled liquid is 250 mg/ml.2.Establish a model of mouse bacteremia induced by E.coli,grouped into two prescriptions: Rescuing Rescuing(Ilex rotunda Thunb+ Cyperus rotundus L.+ Astragalus membranaceus(Fisch.)Bunge.)and Astragalus membranaceus(Fisch.)Bunge.+ Ilex rotunda Thunb Divided into 12 groups,including blank group,model group,negative control group,treatment group(Ilex rotunda Thunb group,Cyperus rotundus L.,Atalantia buxifolia(Poir.)Oliv.ex Benth.,Astragalus membranaceus(Fisch.)Bunge.,Ilex rotunda Thunb + Cyperus rotundus L.,Ilex rotunda Thunb + Atalantia buxifolia(Poir.)Oliv.ex Benth.,Ilex rotunda Thunb + Cyperus rotundus L.+ Atalantia buxifolia(Poir.)Oliv.ex Benth.,Ilex rotunda Thunb + Atalantia buxifolia(Poir.)Oliv.ex Benth.,Xiang Fu + Atalantia buxifolia(Poir.)Oliv.ex Benth.).In the mortality test,the cure rate of Rescuing + Cyperus rotundus L.was the highest,80%,followed by the rescue Ilex rotunda Thunbzhen +Astragalus membranaceus(Fisch.)Bunge.group,the cure rate was 65%;then the Ilex rotunda Thunb + Cyperus rotundus L.+ Atalantia buxifolia(Poir.)Oliv.ex Benth.group,The cure rate is 60%.In the blood routine test,we found that the expression of white blood cells,neutrophils,monocytes,lymphocytes and platelets in the model group decreased,and the platelet volume decreased significantly(P <0.05),and the expression of leukocytes,neutrophils,monocytes,lymphocytes and platelets in the model group was significantly lower than that in the control group(P < 0.05).Compared with the model group,some treatment groups slightly increased the expression of white blood cells,neutrophils,monocytes,lymphocytes and platelets in serum.However,there was no significant difference in the number of leukocytes,neutrophils,monocytes,lymphocytes and platelets in serum of mice(P >0.05).In serum biochemical test,we found that the expression of glutamic-pyruvic transaminase(ALT),glutamic oxaloacetic transaminase(AST),and albumin in the model group was significantly different from that in the blank group(P < 0.05).Compared with the model group,there was no significant difference in all the treatment groups of glutamic-pyruvic transaminase(ALT),(P > 0.05).There was significant difference in glutamic oxaloacetic transaminase(AST),)between Astragalus propinquus Schischkin + Ilex rotunda Thunb group(P < 0.05).For total protein and albumin,there was no significant difference in all treatment groups(P > 0.05).For the valley,There was significant difference in acyl peptidase(GGT),Cyperus rotundus L.and Ilex rotunda Thunb + Atalantia buxifolia(Poir.)Oliv.ex Benth.(P < 0.05).For globulin,the total protein of Ilex rotunda Thunb + Cyperus rotundus L.was significantly different from that of blank group and model group(P < 0.05).Compared with the blank group,the expression of IL-1α,IL-1β,IL-6,IL-10,TNF-α and PCT in the serum of the model group was significantly different from that of the blank group(P < 0.05).Compared with the model group,there was no significant difference in IL-1 α in all treatment groups,but there was significant difference in IL-1β,Astragalus propinquus Schischkin + Ilex rotunda Thunb(P < 0.05).There was no significant difference in all treatment groups of IL-10,(P > 0.05).There was no significant difference in all treatment groups of IL-6,(P > 0.05),but there was significant difference in TNF-α group(P < 0.05),but there was no significant difference in all groups of Astragalus propinquus Schischkin(P < 0.05),but there was no significant difference in all treatment groups(P > 0.05).There was significant difference in PCT,Astragalus propinquus Schischkin(P < 0.05).3.Differential proteomics analysis of the liver of the blank group,the model group,the Cyperus rotundus L.+ Ilex rotunda Thunb group.A total of 5011 proteins were identified in the experiment,of which 4,503 proteins contained quantitative information.If 1.2-fold change threshold and t-test p-value<0.05 are used as the standard,432 protein expressions are up-regulated in the M/B(model group/blank group)comparison group,135 proteins in the quantified protein.The expression is downregulated.In the Y/B(medication group/blank group),the expression of 393 proteins was up-regulated and the expression of 131 proteins was down-regulated.In the Y/M(medication group/model group),the expression of four proteins was up-regulated and the expression of 19 proteins was down-regulated.Glycosaminoglycan binding affects the process of glycoprotein synthesis,decomposition and transport;monocarboxylic acid metabolic process affects the metabolism of three major nutrients,and the down-regulation of the protein means the change of the normal metabolic process of the organism,which indicates that Ilex rotunda Thunb +Cyperus rotundus L.may affect the metabolic process of the substance in mice,thus playing a role in the treatment of bacteremia.
