This study selected the important freshwater aquaculture varieties-Nile tilapia in China as the research object,and discussed the possible regulatory mechanism of hunger stress and autophagy repair through short-term hunger stress and the effect of injection of GSK4112 on the rhythmic expression of Nr1d1 gene and autophagy gene in the liver of Nile tilapia.In this study,real-time fluorescence quantitative PCR(QRT-PCR)was used to determine the rhythmic expression of Nr1d1 gene and autophagy gene in the liver of Nile tilapia,the rhythmic expression was analyzed,the number of autophagy was observed by transmission electron microscope,and the number of autophagy before and after hunger was analyzed.The main findings of the study are as follows:1.Under normal feeding conditions(Control group),the Nr1d1 gene and 12 autophagy genes in the liver of Nile tilapia showed significant circadian rhythm(P<0.30)in Becn1,Gabarapl,Atp6v1 d,Bnip3lb,Map1lc3 a,Map1lc3b,Map1lc3 c,the results showed that the autophagy gene was specifically expressed in the liver of Nile tilapia.Electron microscopy showed that there were multiple autophagy,which indicated that the liver of Nile tilapia was autophagy under normal feeding.This is due to the specific expression of autophagy gene in Nile tilapia liver.2.The significant difference,rhythm,peak phase,amplitude and median of hepatic Nr1d1 gene and autophagy gene were changed before and after 7 days of hunger stress(Experimental group),which showed that hunger altered the original biological rhythm of the gene.The number of autophagy of liver tissue in Nile tilapia increased 7 days after hunger stress was observed by electron microscopy,This indicates that hunger induces the occurrence of tilapia liver autophagy.3.The significant difference,rhythm,peak phase,amplitude and median of hepatic Nr1d1 gene and autophagy gene before and after injection of saline(Control group)and GSK4112(Experimental group)were changed,suggesting that injection GSK4112 changed the original biological rhythm of the gene.The number of autophagy showed decrease in the liver tissue of Nile tilapia was observed by electron microscopy,which showed that GSK4112 inhibited the occurrence of autophagy.From these results,we conclude that hunger stress and injection GSK4112 alter the significant difference,rhythm,peak phase,amplitude and median of hepatic Nr1d1 gene and autophagy genes,and that hunger stress can induce hepatic autophagy,GSK4112 inhibit hepatic autophagy;Short-term hunger stress and injection GSK4112 can alter the original biological rhythms of genes and establish a new balance.
